Version 11-30-2016-1 FAR Part 11—Describing Agency Needs
STATEMENT OF WORK
GENERAL INFORMATION
Title of Project:
DNA-Encoded Chemical Library (DEL) Selection Study
Statement of Need and Purpose:
A list of identified compounds that bind FIP200 protein in a DNA-Encoded Chemical Library screening project .
Background Information and Objective:
We propose to create a national resource of isogenic sets of Alzheimer’s Disease and Related Dementias mutation iPSC lines, complimented by foundational phenotypic datasets from differentiated disease-relevant cells derived from these lines. We anticipate that this resource will substantially accelerate the pace of mechanistic discovery regarding how Alzheimer’s Disease and Related Dementia mutations cause disease.
Period of Performance:
Base period of performance from March 15, 2022- March 14, 2023.
SCOPE OF WORK
General Requirements:
Independently and not as an agent of the Government, the Contractor shall furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government as needed to perform the Statement of Work below.
Specific Requirements:
The Contractor will perform experiments to screen a curated list of fragment and small molecule DNA-Encoded Library collections against of the protein FIP200 under both normal and stringent selection conditions. The results of the screening will be returned to the Government in the form of a blinded selection report on the identified compounds that bind FIP200.
Vendors technology for library screening will include:
• Biologically relevant hits to protein of interest that are pharmaceutically relevant.
• Strict filtering to remove promiscuous binders and known toxic substructures, while mantaining an average Tanimoto similarity of below 0.2
• Vendor’s library should be built on two DNA strands which anneal together to present structural pairs to the target. These structural pairs shall constantly reshuffle in solution to generate optimized binding pairs.
• Vendor shall use Path-Coding system for encoding/decoding DNA barcodes for libraries allowing for built in error detection and correction in library sequencing.
• Vendor shall use methods allowing for seemlessly integration with data previously obtained from Government’s DEL screening of the protein FIP200 under both normal and stringent selection conditions.
LEVEL OF EFFORT:
Vendor staff will provide the results via reporting transfer through email.
GOVERNMENT RESPONSIBILITIES
The government will provide starting recombinant protein material to the vendor as needed.
DELIVERY OR DELIVERABLES
A report containing information on the identified hit compounds, including their enrichment values, physicochemical metrics, and chemoinformatic/clustering data shall be delivered to the Government.
OTHER CONSIDERATIONS
Travel:
The principal place of performance is the vendor’s work site. Travel for contractor employees will not be necessary.
Key Personnel:
Vendor will provide a dedicated project manager to the government as a point of contact for the services.
Information System Security Plan:
Not Applicable
Data Rights:

Section 508—Electronic and Information Technology Standards:
The contractor shall comply with Section 508 of the Rehabilitation Act (29 U.S.C. § 794d), as amended by the Workforce Investment Act of 1998 (P.L. 105-220), August 7, 1998.
