PURCHASE DESCRIPTION
1. Statement of Need and Purpose:
NCATS needs human alveolar and tracheobronchial tissues in a ready-made format for drug testing/viral infection for NCATS ETB SARS-CoV-2 project.
2. Background Information and Objective:
Lung is one of the first organs where respiratory viral infection occurs, and severe diseases have been reported to occur in lung parenchyma. Thus, physiologically relevant in vitro lung models are highly demanded to help finding treatments and studying host responses to virus infections and drug treatments. We have established protocols for barrier tissue models with vascularization using 3D bioprinting of required cellular components in a transwell format.
The acquisition of alveolar and tracheobronchial tissues will provide the program an extensive opportunity to validate potential anti-SARS-CoV2 compounds, as well as to characterize host response in a 3D lung model to SARS-CoV2 and other high impact respiratory viruses.
3. Generic Name of Product: Human alveolar and tracheobronchial tissues.
4. Purchase Description: COVID-19/Coronavirus – the alveolar and tracheobronchial tissues in a ready-made format for drug testing must be brand-name or equal to the following:

5. Salient characteristics: The brand equal items must meet the following salient characteristics:
• Must come in a ready-made format for drug testing/viral infection.
• Must be able to recapitulate the in vivo phenotypes of barrier, mucociliary responses, infection, toxicity responses and disease.
• Must exhibit human relevant tissue structure and cellular morphology with high uniformity and reproducibility.
• Must be suitable for a host-pathogen interaction study and antiviral therapeutic screening.
• The epithelium must exhibit functional tight junctions and a robust barrier that remains stable for long lasting studies.
