About this file

This document is a purchase description for a tunable femtosecond laser system to replace damaged equipment for the National Institute of Neurological Disorders and Stroke. The purchase description specifies a Coherent NA, Inc. Chameleon Discovery NX laser system or equal that can deliver high peak power directly to the sample plane using built-in dispersion precompensation. The system must have outputs at 800nm and 1040nm with short pulses and high power to enable multimodal microscopy for in-vivo imaging applications when integrated with an existing LSM980 multiphoton microscope. Delivery within 112 days of order is required to the specified NIH building. An OEM certified technician must perform setup, installation, and training within 30 days of delivery. The purchase description also includes a trade-in of a damaged Coherent Ultra II/Vision laser system.

The related federal contract opportunity is a solicitation from the National Institutes of Health National Institute on Drug Abuse seeking the tunable femtosecond laser system specified in the purchase description.

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

TITLE: Coherent NA, Inc., Chameleon Discovery NX (Part #1404200) or Equal with Trade-In

1. Statement of Need and Purpose:

The purpose of this requirement is to procure one (1) Chameleon Discovery NX or equal for the National Institute of Neurological Disorders and Stroke (NINDS) for replacement due to damage by the flood in Building 35. This tunable femtosecond laser system is designed for multiphoton microscopy and shall be used to provide dual laser power to the NINDS government-owned LSM980 multiphoton system used to take live imaging of Drosophila brain with fluorophore probe to obtain deeper and brighter than usual images when NINDS researchers collect in-vivo data, which is used for analysis dynamic change at the early dendrite development stage.

2. Background Information and Objective:

The NINDS, which is an Institute at the National Institutes of Health (NIH), mission is to seek fundamental knowledge about the brain and nervous system and to use that knowledge to reduce the burden of neurological disease for all people.

The NINDS Dendrite Morphogenesis and Plasticity Section (DMPU) applied the Drosophila larval visual circuit and investigates the structural and functional plasticity changes to find the mechanisms underlying the neuronal development. The DMPU is using in-vivo live imaging with the aid of a multiphoton scanning system to study different candidate genes. The primary approach is to understand molecular path to regulate dendrite morphogenesis and structural plasticity, and learn the consequent function change due to deficits on homeostatic neuronal plasticity.

On December 24 and 25, 2022, NIH Building 35 equipment malfunctioned and caused flooding that effected multiple NINDS laboratories and offices including the DMPU.

The NINDS DMPU requires a tunable femtosecond laser system designed for multiphoton microscopy to replace an older system which sustained water damage in the recent flood. The requiring office identified Coherent NA, Inc., as a potential vendor that offers a tunable femtosecond laser system known as the Chameleon Discovery NX.

For this procurement, offerors must take into consideration the following trade-in equipment when providing a quote to offer a discount to the purchase price of the new property in exchange for the old, similar property (refer to section 10 of the purchase description below).

Acquiring a tunable femtosecond laser system will enable DMPU investigators to renew its ongoing research studies, which have been halted because of the flood damage. Its investigators can obtain deeper and brighter than usual images when collecting in-vivo data from the use of the dual laser power of the system integrated with the government-owned LSM980 multiphoton system to take live imaging of Drosophila brain with fluorophore probe. The data is then used for analysis dynamic change at the early dendrite development stage.

To reach the most function of the NINDS government-owned LSM 980 multiphoton system, the Chameleon Discovery NX or equal will enable a high power laser for multiphoton microscopy to make brighter and deeper live imaging possible in the DMPU’s new and ongoing research studies.

3. Generic Name of Product:

Tunable Femtosecond Laser System

4. Purchase Description:

For reference purposes, the brand-name tunable femtosecond laser system that meets the need of this requirement is as follows:

Part Number

Description Manufacturer Quantity

1404200 Chameleon Discovery NX Coherent NA, Inc. One (1)

5. Salient Characteristics:

The significant, essential physical, functional, performance, or material element(s) that are the Government’s minimum acceptable standards for an equivalent system to meet the government’s objective are as follows:

The tunable femtosecond laser system shall:

• be fully automated for hands free operation to delivers high peak power directly to the sample plane using built in, high dynamic range dispersion precompensation for the tunable output and have the combination of short pulses (100fs at900nm), high average power (> 3.6 W at 800nm) and widest tuning range (660-1320nm) to give brighter and deeper images for in-vivo imaging applications.

• have a second output at 1040nm, with high power (>3500mW) and short pulses

(140fs) to further enable multimodal capability for dual color excitation and activation of optogenetics probes. The two outputs shall have a phase locked to enable techniques including CARS, SRS and 2P wavelength mixing.

• have a graphical user interface (GUI) compatible with PC Windows and be controlled directly via a RS232 or USB interface located on the rear of the laser head.

• exact industrial standards, using HASS and HALT methodologies.

• have a single-phase AC power and include:

o compact laser head with integrated dispersion precompensation and light loop power control for long term stability;

o power supply unit with shutter control and interlocks;

o closed-loop water chiller; and o MRU-X1 closed-loop air re-circulator for ensuring clean integrity of the ultrafast laser cavity.

• shall be capable of integrating with the government-owned LSM980 multiphoton system.

• the system shall be warranted, and the Contractor shall warrant its equipment against defects in material, fabrication, or workmanship for a minimum of three

(3) years effective from the date of installation.
