About this file
This document is a purchase description for an automated purification system. The National Institute of Neurological Disorders and Stroke Proteomics Core Facility requires an automated system to process protein samples in order to improve throughput and reproducibility compared to current manual methods. The system must meet specified requirements including processing samples in 96-well plates with volumes from 10 microliters to 5 milliliters using magnetic beads, accommodating 24 to 96 samples simultaneously, including temperature control and integrated software. Delivery, installation, calibration and three days of on-site training must be completed within twelve weeks of order receipt. A minimum one-year warranty is required along with ongoing technical support services. The related federal contract opportunity was issued by the Department of Health and Human Services National Institutes of Health National Institute on Drug Abuse for this automated purification system.

PURCHASE DESCRIPTION
Brand name or equal: KingFisher Apex Purification System
1. Statement of Need and Purpose:
The National Institute of Neurological Disorders and Stroke (NINDS) Proteomics Core Facility (PCF) requires a brand name or equal KingFisher Apex Purification System. This instrument will be used to automate cleanup of protein samples and represents a significant improvement over the current manual process.
2. Background Information and Objective:
The National Institute of Neurological Disorders and Stroke (NINDS) Proteomics Core Facility (PCF) provides protein identification, protein post-translational modification mapping, and protein quantitation services for NIH-wide investigators. The facility is also utilized for collaborations involving various complicated protein quantitation strategies.
Over the past several years, identifying protein binding partners via labeled or label free quantitation workflows has been the most requested services by PCF users. Most samples contain detergent which is not compatible with the downstream mass spectrometry analysis.
Traditionally, protein precipitation is used to remove the detergent from the protein samples.
The method has relatively poor sample cleanup and low protein yield. The recently developed S-trap method involves more than 20 manual steps. It is so time consuming that it becomes the main reason for longer turnaround time.
The required instrument automates cleanup of protein by moving magnetic beads, not liquids, which enables the removal of detergent with consistent protein extraction and high protein yield. Not only can this instrument process a much greater number of samples simultaneously than is possible manually, it will also reduce errors and increase reproducibility, which is crucial for the success of labeled or label free quantitation services used by majority of the PCF users.
3. Generic Name of Product:
Automated Purification System
4. Purchase Description:
Brand Name or Equal Benchmark: ThermoFisher Scientific KingFisher Apex Purification System Manufacturer’s Name: ThermoFisher Scientific
5. Salient characteristics Contractor shall provide the NINDS PCF an automated purification system that meets or exceeds the following requirements:
• An automated system for the separation, purification and extraction of biological samples, including cells, organelles, proteins, peptides and nucleic acids..
• Uses a unique approach in which superparamagnetic particles or magnetic beads are moved instead of transferring liquids.
• Purification and processing steps are performed in 96 deepwell plates, 96 KF plates, 96 PCR plates, or 24 deepwell plates or selected screw cap storage tubes with dimensions that match the configuration of tip combs that cover the magnetic rods.
• Can accommodate processing volumes 10 µl - 5ml.
• Processes up to 24 or 96 samples at a time and can be in variable amounts chosen by the end user.
• The system includes turntable with 8 plate positions.
• The system has up to 2 snap-on heat blocks for applications requiring temperature control in the range of +4 – 100 ºC.
• The system includes display on loading position with capability to show status for consumable loading.
• The system allows to leave samples in desired temperature after completion of the run..
• The system includes ~10 inch touch screen with integrated BindiX Software with capabilities to control all operational functions on the unit as well as retrieve protocols from Connect.
• The integrated BindIx PC software allows user to create, modify, save and run protocols without the need for an attached computer
• The system includes integrated bar code reader capable of reading 1D bar codes
• The system allows input and use of up to 2 magnet heads.
• Two magnet head types of the system allow use of 2 different tip combs according user selection.
• The system creates run reports which can be saved into instrument, USB stick.
• The system includes short term memory allowing user to continue the run after power breakage from the point it was stopped.
• The system is an open platform capable of using any manufacturer’s magnetic particle based kits and associated protocols and is not limited to running only one kit supplier’s chemistry systems
