About this file

This document outlines the purchase description for a federal contract solicitation seeking two Digidata patch clamp amplifiers and associated software from the National Institutes of Health National Institute on Drug Abuse. The solicitation requests equipment that meets or exceeds specifications for a Digidata 1550B data acquisition system and pClamp 11 software from Molecular Devices to replace items damaged in a flood. The National Institute of Neurological Disorders and Stroke Laboratory of Circuits, Synapses, and Molecular Signaling requires the two patch clamp amplifiers for single channel, whole cell patch, and extracellular electrophysiology recordings from neurons and cells. The amplifiers must support current clamp and voltage clamp modes with specific holding current, potential, gain, compensation, and noise parameters.

PURCHASE DESCRIPTION

Brand Name or Equal: Two Digidata patch clamp amplifiers and software

1. Statement of Need and Purpose:

The purpose of this requirement is to acquire two new brand name or equal Digidata 1550B digitizers and pClamp 11 electrophysiology analysis software for electrophysiology and pharmacological experiments at the National Institute of Neurological Disorders and Stroke (NINDS). These items are extensively used in experiments at many National Institutes of Health (NIH) laboratories. NINDS’ previous digitizers were damaged in a flood at NIH Building 35 in December 2022.

2. Background Information and Objective:

The NIH is an agency of the Department of Health and Human Services and the Federal focal point for health research. Its mission is to seek fundamental knowledge about the nature and behavior of living systems and apply that knowledge to enhance health, lengthen life, and reduce illness and disability.

The NINDS Laboratory of Circuits, Synapses, and Molecular Signaling (LCSMS) studies fundamental issues of synaptic transmission, neuronal excitability, neuromodulation, and neural circuit activity using animal models (principally, the mouse). A major line of research employs patch clamp electrophysiology, fluorescence imaging, and optogenetic stimulation in brain slices. For this research, the LCSMS requires two (2) low-noise patch clamp amplifiers, suitable for single channel recording and whole cell patch recording from neurons in brain slices. The amplifiers must also be compatible with extracellular recording.

These items are needed to record electrical activity from neurons and heterologous cells expressing ion channels. Both hardware and software are routinely used for acquiring and analyzing electrophysiological activity in several labs at NIH, including NINDS’, and worldwide.

3. Generic Name of Product:

Analog-to-digital single converter (digitizer), data acquisition and analysis software

4. Purchase Description:

• Brand name or Equal benchmark: Digidata 1550B4 data acquisition w/4x

HumSilencer

• Manufacturer’s name: Molecular Devices LLC

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

5. Salient characteristics:

The LCSMS requires two (2) patch clamp amplifiers for single channel, whole cell patch, and extracellular recordings.

The equipment must meet or exceed the following specifications:

o Compatible with the Government’s existing Digidata 1550B data acquisition system and pClamp 11 software (manufactured by Molecular Devices).

o Actively cooled headstage for low noise recording: 0.06 pA rms (5 kHz), 0.13 pA rms

(10 kHz).

o Current clamp and voltage clamp recording modes.

o Holding current range ±200 nA.

o Holding potential range ±1000 mV.

o Output gain (mV/pA) 0.5 to 500 pA.

o Capacitance compensation.

o Seal test functionality.

o Telegraph outputs for gain, filter, and capacitance.

o Must be suitable for single channel recordings which have strict noise requirements

(<0.15 pA RMS at 10 kHz sample in voltage clamp).

o Must be suitable for whole cell patch recording (both current clamp and voltage clamp) from neurons in brain slices.

6. Quantity: Two (2) systems

7. Delivery Terms: The Contractor shall deliver the equipment within 30 days after receipt of order to the following location:

NIH Building 35 35 Convent Drive Rm 3C-1004 Bethesda, MD 20892

Delivery must be Freight on Board (FOB) Destination and include inside delivery. All equipment shall be shipped/delivered in coordination with the Contracting Officer’s Representative (COR).

8. Warranty: Contractor shall warrant its equipment against defects in material, fabrication, or workmanship for a minimum of 1 year from the date the Government accepts delivery of the items. This includes replacement of damaged devices.
