Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

Brand name or equal: Seahorse XF Pro Complete w/Normalization

1. Statement of Need and Purpose:

The purpose of this requirement is to acquire a brand name or equal Agilent Seahorse XF Pro Complete w/Normalization. This requirement will support the research at the Synaptic Function Section (SFS) of the National Institute of Neurological Disorders and Stroke (NINDS) Division of Intramural Research (DIR).

The SFS’ previous XF analyzer was damaged in flooding to NIH Building 35 on December 24-25, 2022. Replacing the damaged analyzer with a new system is the best interest of the Government for continuation of science, resuming SFS’ ongoing and long-term research activities, and for training postdoc fellows and graduate students.

2. Background Information and Objective:

Research at the SFS aims to investigate axonal mitochondrial energy metabolism and lysosomal degradation capacity in neurons. It has applied cutting-edge live and super-resolution imaging assays and energy metabolism analysis in a multidisciplinary system analysis using cultured neurons and genetic mouse models combined with human isogenic iPSC-derived neurons harboring disease-linked mutations. SFS’ expected research contributions, will be significant because these studies will advance our knowledge of fundamental cellular and energy metabolic processes affecting synaptic transmission, CNS regeneration, adult neurogenesis, and aging-associated neurodegeneration, and thus directly align with the mission and strategic plan of NINDS.

A major part of the research activities within the SFS critically depends on XF analyzers, which is a technology platform used for measuring cell energy metabolism in live cells in real time. XF analyzers simultaneously measure the two major energy producing pathways of the cell – mitochondrial respiration and glycolysis – in living cells, in a microplate, and in real time. Researchers are utilizing this technology for faster, better, and more accurate measurements of cellular metabolism. Specifically, by using this technology, the SFS can simultaneously measure oxygen consumption rate (OCR) and extracellular acidification rate (ECAR) of living cells. Using these measurements, glycolytic Protein Efflux Rate (PER) is easily calculated as an accurate measurement of glycolysis rates in real time. Quantitative ATP production rates can also be calculated in real time using well-validated algorithms to obtain mito-ATP and glycol-ATP production rates, as well as total ATP.

Since its acquisition in 2018, this Agilent Seahorse XFe96 Analyzer has been a key metabolic tool for the SFS’ daily research activities in studying energy metabolism in live neurons in the past four years. By using this analyzer, the SFS has published research in three papers of high-profile journals. To ensure continuity of research, it is critical that the replacement instrument is equivalent to the previous analyzer and fully compatible with all equipment that the previous analyzer was used in conjunction with.

3. Generic Name of Product:

Extracellular Flux (XF) Analyzers

4. Purchase Description:

Brand name or equal benchmark: Seahorse XF Pro Complete w/Normalization Manufacturer name: Agilent Technologies

5. Salient characteristics

General Requirements:

• Simultaneously measures oxygen consumption rate (OCR) and extracellular acidification rate (ECAR) of living cells. Using these measurements, glycolytic Proton Efflux Rate (PER) is easily calculated as an accurate measurement of glycolysis rates in real time. Quantitative ATP production rates can also be calculated in real time using well-validated algorithms to obtain mito-ATP and glycol-ATP production rates, as well as total ATP.

• Performs a non-invasive measurement that does not require the addition of dyes, labels, or reporters.

• Cells/microplate remain viable following an assay and may be used for another assay.

• Measurements may be repeated to measure kinetic responses over several hours without causing unhealthy hypoxic or acidic conditions.

• No cleaning is required. All the parts that are in contact with the cells, media, or compounds are disposable.

• Measures adherent cells without requiring trypsinization; suspension cells can be used in conjunction with Cell-Tak® cell and tissue attachment medium.

• Analyzer can be interfaced with the Biotek Cytation 1 and Citation 5 for a validated, unified normalization solution.

Specific Requirements:

• Instrument creates transient microchamber, makes optical measurements, and performs compound injections.

• Up to four test compounds may be added automatically to each well during the assay.

Measurements may be performed before and after each compound is added.

• Typically requires only 1x104 to 1x106 cells per 24-well.

• Temperature controlled measurement chamber is maintained at 37°C ± 0.5°C;

alternative temperatures supported.

• OCR and ECAR data are simultaneously calculated and displayed in real time. OCR and ECAR enable for calculations of glycolytic PER and ATP production rates (mito- ATP and glycol-ATP).

• Software provides an intuitive experience across all steps of the assay workflow, from designing experiments to analyzing & reporting results. Data can easily be extracted & exported to Microsoft Excel & GraphPad Prism file formats for additional analysis needs.

• Optical sensors do not consume oxygen during the measurement and are not in contact with the cells.

• Optical sensors are not affected by compound or intracellular dye fluorescence.

• Oxygen sensor peak absorption = 530 nm (green). Oxygen peak emission = 650 nm

(red). pH sensor peak absorption = 470 nm (blue). pH sensor peak emission = 530 nm (green).

• Light Emitting Diodes (LEDs) are used as the monochromatic excitation source. LEDs are operated at very low excitation energy densities to prevent detectable levels of photobleaching.

• Narrow band pass filters control spectral adherence.

• The precision-molded enclosures on analyzers have specialty coatings and gaskets to shield the electronics and sample from ambient light, as well as block radiated and emitted electromagnetic interference.

• Rates are automatically calculated and reported every 5-8 minutes.

• Quality-controlled kits provide standardized metabolic measurements in an easy-to-use format.

• Assays are non-invasive, multiple measurements may be made to monitor samples over several hours. Measurement-to-measurement within the same well demonstrates a coefficient of variation of 5% approximately equivalent to the instruments’ background noise. Total variability between wells from all sources (instrument background plus biological variability due to cells) for the average trained user is <20%.

• A supported normalization solution interfaces analyzers with BioTek Cytation 1 and Cytation 5 imagers to apply cell count numbers directly to XF data, enabling validated, plate-to-plate, day-to-day, and well-to-well comparisons.
