About this file

This document is a Request for Quotations (RFQ) for a commercial item - a Single nuclei multiome sequencing kit, including nuclei partitioning gel beads, and cell partitioning chips. The National Institute of Neurological Disorders and Stroke (NINDS) requires these reagents to generate ATAC-seq and RNA-seq libraries from individual cells for ongoing research in the Molecular Neurobiology Section.

The RFQ specifies the required product characteristics, quantity, delivery timeframe of 30 days, and applicable FAR clauses. It solicits quotes with a submission deadline of July 29, 2024. The acquisition will be a firm-fixed price purchase order awarded to the responsible offeror whose quote conforming to the solicitation will be most advantageous to the Government, considering technical capability, price, and past performance. The associated NAICS code is 334516 with a small business size standard of 1,000 employees.

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

Title: Single nuclei multiome sequencing kit, including nuclei partitioning gel beads, cell partitioning chips compatible or equal

1. Statement of Need and Purpose: The Molecular Neurobiology Section (MNS) within the National Institute of Neurological Disorders and Stroke (NINDS) investigates mechanisms of neural regeneration after injury using single cell/single nuclei sequencing and has a need for reagents and chips for capturing single nuclei using a 10x Chromium X nuclei partitioning device and generating multiome (open chromatin and 3’ gene expression) libraries from the same individual cells. The reagents currently in the lab need to be resupplied.

These reagents will be used to generate both ATAC-seq and RNA-seq libraries from the same individual cells. These libraries are necessary for continuing ongoing experiments in the lab and need to be consistent with these ongoing experiments.

2. Background Information and Objective: The National Institutes of Health (NIH) is an agency of the Department of Health and Human Services and the Federal focal point for health research. Its mission is to seek fundamental knowledge about the nature and behavior of living systems and apply that knowledge to enhance health, lengthen life, and reduce illness and disability.

The NINDS, which is an Institute within the NIH, seeks to reduce the burden of neurological disease through supporting and conducting basic, translational, and clinical research on the healthy and diseased nervous system, fostering the training of investigators in the basic and clinical neurosciences, and seeking better understanding, diagnosis, treatment, and prevention of neurological disorders. to find out as much as possible about the brain and nervous system in order to lessen the burden of neurological disease.

The main goal of the MNS is to identify the signaling pathways important for neural regeneration after injury and currently uses the mouse olfactory epithelium, which has robust regenerative capacity, as a primary model. A primary approach to understanding mechanisms of neural regeneration is to sequence the genes that are expressed in in individual cells and the regions of open chromatin, which correlate gene regulatory elements, in individual nuclei. The reagents needed will allow the MNS to identify specific genes and regulatory elements in the same individual cells of the olfactory epithelium of mice before and after injury.
