(vi) Statement of Need:
Background:
The NIAMS Genomic Technology Section is charged with providing genome-scale, high throughput sequencing services for researchers within the institute.
The Core Facility, like most other Institutes at NIH and sequencing centers around the world, adopts new emerging technologies in order to maintain the leading edge on genomics and epigenetic research areas.
Currently, NIAMS core facility uses the 10X Genomics Chromium Controller Single Cell Platform that is a molecular barcoding and analysis suite that delivers cell-by-cell 3’ counting of mRNA transcripts for many tens of thousands of cells per run. The platform supports a broad range of applications, including single-cell spatial transcriptomics, cancer-cell transcriptomics and cell-type identification and discovery. Because the platform works with short read sequencers it is configured to be compatible with NIAMS Illumina sequencing systems and will be a very important tool for our researchers to continue performing genomic and epigenetic research projects.
The Chromium Controller Single-Cell Platform provides high throughput reagent delivery using a microfluidic chip. The 8-sample cartridge is loaded with the gel beads, a 3’ counting reagent, a cell suspension, and an oil-surfactant solution.
Reagents and cells are combined at a microfluidic junction where gel bead-containing emulsions (GEMs) are generated. GEMs are created in such a way that single cells are partitioned and >90% of reactions contain a unique cell-linked molecular barcode. The simple workflow creates up to 100,000 GEMs per channel in a 10-20 minutes instrument run time.
To support the research of NIAMS Investigators, there is need to purchase reagents compatible with this system. The Chromium Controller Single Cell Platform uses the Chromium Single Cell Library & Gel Bead Kit, the Chromium Single Cell ATAC Library & Gel Bead Kit, Library Construction, Single Cell and Index kits to produce epigenetic information of thousands of cells of up to eight different cell populations. These tools will support several research projects carried out to better understand the genetic determinants of autoimmune, auto inflammatory and musculoskeletal diseases and gain insights into the pathogenesis of genetic and immunologic diseases at a single-cell level.
The work of the NIAMS Genome Analysis Core Facility depends heavily on sequencing of DNA Libraries from tissues, cell lines and single cell populations from various species. The purchase of these reagents is needed to operate and conduct experiments in the Chromium Controller Platform; it is fundamental to access the cutting-edge technology in the area of single-cell epigenetic research that enable discovery of gene expression dynamics and molecular profiling of individual cell types. This acquisition fulfills the mission of the Core Facility on meeting the demand for new tools to support NIAMS research.
