Olink® Proximity Extension Assay Support Services
Division of Translational Toxicology (DTT)
Research Triangle Park, NC
NOTE: This is a Brand Name or Equal solicitation in accordance with (IAW) FAR Subpart 11.104-Use of brand name or equal purchase descriptions and FAR provision 52.211-6:
Brand Name or Equal. The vendor shall implement the proximity extension assay (PEA) technology developed by Olink® on multiple pilot studies for assay sensitivity, specificity and high throughput capabilities for applications of cancer and environmental exposure biomarker discovery. Section 2.2 provides a general description of those salient physical, functional, and/or performance characteristics of the brand name Olink® PEA that an “equal” item/solution must meet to be acceptable for award.
1.0 INTRODUCTION / OVERVIEW
The Division of Translation Toxicology (DTT) is an interagency program whose mission is to evaluate agents of public health concern by developing and applying tools of modern toxicology and molecular biology. The program maintains an objective, science-based approach in dealing with critical issues in toxicology and is committed to using the best science available to prioritize, design, conduct, and interpret its studies. To that end, DTT is continually evolving to remain at the cutting edge of scientific research and to develop, validate and apply new technologies.
1.1 Background
Targeted multiplex immunoassays are currently being applied as a biomarker tool for disease detection and prognosis in the clinical setting. Lessons learned clinically can be introduced, applied, and translated to predict similar disease outcomes in exposure biology and toxicology.
High throughput proteomic assays can be used to help interpret hazard and risk for multiple different phenotypes such as coronary vascular and neurological disorders, cancer, inflammation, and organ damage. As a tool, it can be used to screen, predict, and further investigate these phenotypes with translation to the human population, especially in the wake of environmental accidents. From a non-invasive, blood liquid biopsy (whole blood), real time circulating protein expression can be obtained and provide acute and chronic disease information. These data can be applied to disease and biologic process biomarker discovery, predictive dose and disease effects generated by computational toxicology.
The Vendor shall implement the proximity extension assay (PEA) technology developed by Olink® (OR EQUAL) on multiple pilot studies for assay sensitivity, specificity and high throughput capabilities for applications of cancer and environmental exposure biomarker discovery. A major premise of this project is that protein expression signatures underlie organ toxicity and the development of chemically-induced pathology and tumors. Alteration and activation of critical genes and other regulatory pathways enhance or repress gene expression leading to toxicity or cancer susceptibility. Protein expression signatures can be interpreted as molecular biomarkers which can precede or inform histopathology and other measures of toxicity.
The vendor shall provide knowledge and expertise in proteomics utilizing the Olink® PEA technology platform (OR EQUAL) on liquid biopsy (serum and plasma) samples.
1.2 Scope
The vendor shall perform all tasks required to complete and deliver reliable Olink proximity extension assays as delineated in Section 2.0.
2.0 REQUIREMENTS
The vendor shall be authorized, qualified, and have experience performing the Olink® PEA (OR EQUAL). The vendor shall follow protocols set forth by Olink® (OR EQUAL) to run blood or urine samples DTT shall be contacted at any time of the process if there are problems with samples or completion of the assay run. Regular status updates shall be provided to DTT at the completion of steps associated with the PEA. This includes receipt of samples, completion of sample preparation, the panel que for PEA, assay completion and data transfer.
