Requirement The U.S. Food and Drug Administration (FDA), Center for Biologics Evaluation and Research (CBER), requires a contractor capable of providing, housing and evaluate up to 24 vaccinated baboons, house and evaluate up to 12 pertussis-convalescent baboons (provided by CBER) for up to five years. The contractor will also be required to collect samples from 12 age-matched, unvaccinated animals selected from contractor’s colony.
Background:
Pertussis is an acute, highly contagious, potentially life-threatening respiratory disease, caused by the Gram-negative bacterium Bordetella pertussis. The underlying factors leading to the failure of public health measures to control pertussis disease using currently licensed vaccines are poorly understood. A more complete understanding of this disease and of the mechanisms of vaccine-mediated protection is a critical public health need. Currently, the baboon model of pertussis, developed in the laboratories of the Food and Drug Administration (FDA) Center for Biologics Evaluation and Research (CBER), is the only animal model of pertussis that recapitulates the disease as observed in human patients. The use of this animal model to study the Immune Response to Infection and Vaccination requires the conduct of a multi-year study following immune responses in up to 48 animals.
Minimum Technical Capabilities & Requirements:
• Contractor shall provide 24 - 5-7 month-old Bordetella-free Baboons into study within twelve months of award of contract
• Contractor shall have at least 12 age-matched unvaccinated control animals available at time of semi-annual sampling o Age-matched, unvaccinated animals do not have to be the same animals at every sampling although that is optimal mailto:howard.nesmith@fda.hhs.gov
• Contractor shall have facilities and procedures that protect enrolled animals from Bordetella exposure for the duration of the study o Facilities shall be subject to inspection and approval by the CBER Principal Investigator
• Contractor facility shall be required to have and maintain AAALAC accreditation
• All animals enrolled in the study shall be owned by the Contractor and overseen by the
Contractor’s (American Association for Laboratory Animal Science) IACUC
• Contractor shall communicate to CBER any program deficiencies, that impacts the care and use of the study animals, identified by their IACUC or external site visitors within 30 days
• Contractor shall vaccinate three times, two-months apart, with FDA provided acellular or whole cell pertussis vaccines (See Attached Appendix A)
• Contractor shall draw blood and perform nasopharyngeal washes on the 24 animals in the vaccine groups upon enrollment into the study (See Attached Appendix A) o Nasopharyngeal washes shall be plated by the contractor to confirm lack of Bordetella colonization and sera shall be sent to CBER laboratories for serological analysis to confirm lack of Bordetella-exposure o Sera and nasopharyngeal wash from vaccinated animals shall be provided at time of enrollment
• Contractor shall be required to perform all procedures as described in Appendix A
• Animals shall be maintained for up to five years under conditions that will minimize risk of exposure to Bordetella
• Contractor shall commit up to five year-duration of study o CBER may terminate the study early if endpoints are met o The study may be extended at the conclusion of the five-year period upon completion of a new “follow-on” contract
• Contractor’s facility shall be subject to inspection by the CBER Principal Investigator to evaluate adequacy of proposed measures to prevent exposure of study animals prior to award of contract

Contractor Performance Evaluation(s) In accordance with Federal Acquisition Regulation (FAR) 42.15, FDA will complete annual and final contractor performance evaluations. Annual evaluations will be prepared to coincide with the anniversary date of the contract. Additional interim performance evaluations may be prepared at the CO’s discretion, as necessary. Final performance evaluations will be completed upon contract expiration.
FDA will utilize the Contractor Performance Assessment Reporting System (CPARS) in order to execute annual and final contractor performance evaluations. CPARS is a secure Internet website located at https://www.cpars.gov/main/cparsmain.htm. FDA will register the contractor in CPARS upon receipt of the name and email address of two (2) individuals who will be responsible for serving as the Contractor’s primary and alternate CPARS contacts. Once FDA registers the contractor in CPARS, the Contractor will receive an automated CPARS email message which contains User IDs and instructions for creating a password.
Once a performance evaluation is issued, the Contractor’s primary and alternate CPARS contact will receive an email instructing them to logon to CPARS in order to review the performance evaluation. The Contractor has 14 calendar days from the date of performance evaluation issuance in which to review the evaluation. If the Contractor is in agreement with the performance evaluation outcome, the evaluation becomes final. Should the Contractor be in disagreement with the performance evaluation outcome, rebuttal comments must be submitted via the CPARS within 30 days from date the evaluation was issued by FDA. Any disagreement between the CO and the Contractor will be referred to a contracting official one level above the CO, whose decision will be final.
Copies of each performance evaluation and contractor responses, if any, will be retained as part of the official contract file and will be used to support future award decisions. Evaluations will also be stored for a 3 year period (6 years for construction) in the Past Performance Information Retrieval System (PPIRS) at www.ppirs.gov.
https://www.cpars.gov/main/cparsmain.htm
Contractors may obtain CPARS training material and register for on-line training at https://www.cpars.gov/main/cparsmain.htm. There is no fee for registration or use of the CPARS system.
