(vi) Statement of Need:

Background:

The National Institutes of Health (NIH), a part of the U.S. Department of Health and Human Services, is the nation’s medical research agency — making important discoveries that improve health and save lives. The mission of the (NIAMS) is to support research into the causes, treatment, and prevention of arthritis and musculoskeletal and skin diseases; the training of basic and clinical scientists to carry out this research; and the dissemination of information on research progress in these diseases. The NIAMS Flow Cytometry Facility (FCF) provides research support to members of NIAMS IRP and its collaborators.

Serving all NIAMS investigators, both research and clinical, the FCF operates four flow cytometry cell sorters as well as oversees the use of six analyzers including a 5-laser, 64+3 channel Cytek Aurora spectral cytometer. The FCF supports approximately 120+ users seven days a week. Cell sorting services are provided Monday through Friday. Each instrument supports multiple experiments each day, and each instrument is fully utilized.

Objective:

The purpose of this requirement is to provide NIAMS investigators with state-of-the-art immunological assays instruments to able to develop a higher parameter staining panels using the existing 40-color Cytek Aurora spectral cytometer (purchased in 2018), and to able to easily sort and discover populations using compatible optical detectors and cell sorters

Scope:

The contractor shall provide and install an instrument with features brand name or equal to the Cytek Aurora-CS 5L Full Spectrum Sorter and Cytek Aurora 5- Laser UV/V/B/YG/R Systems as listed in section 16(v), including system software and 12 months manufacturer’s full warranty including parts and labor.

Minimum Technical Requirements

• Be a stand-alone multi-parameter spectral analyzing cytometer and cell sorter system, and include all required acquisition and analysis software

• Able to simultaneously discriminate up to 40 highly overlapping fluorochromes using Coarse Wavelength Division Multiplexing (CWDM) detection arrays.

• Capable of full spectrum unmixing

• Include three (3) light scatter parameters

• Incorporate side scatter detection laser (violet);

• Capable of small particle resolution down to 0.1 microns

• Incorporate five (5) spatially separated lasers as follows: UV, 355nm at

20mWatt minimum; violet, 405 nm at 100mWatt minimum; blue; 488 nm at 50mWatt minimum; yellow green, 561nm at 50mWatt minimum; red, 640 nm at 80m Watt minimum

• Accommodate a variety of sample tube formats for manual sample delivery

• Cell sorter must:

o include sample and collection tube cooling and heating o be capable of multi-well plate sample delivery o be capable of variable sample handling and variable sort collection o be capable of handling 70um and 100um nozzle sizes o include primary and secondary biosafety controls to include (spell out) High-Efficiency Particulate Air (HEPA) filter (primary) for sort chamber aerosol management and Biosafety Class 2 Type A2 cabinet (secondary) tested to major WW biosafety standards o include 1 additional copy of acquisition/analysis software, QC and consumable parts kits
