Performance Work Statement (PWS) Design and Content

1.0 Introduction (Provide a brief description of what you are buying).

JuLI™ Stage is a fully automated real-time cell imaging system that directly acquires cell images from numerous cell culture plates (6 up to 384 wells) and dished in an incubator.

2.0 Background (Provide any historical information necessary to understand how and why the requirement evolved and where it is headed. Include appropriate information to help the reader/contractor understand the requirement).

Imaging of living cells and tissues is instrumental in revealing cellular dynamics and functions. Live cell imaging enables the observation of dynamic processes across entire cell populations, individual cells, or even at the subcellular level. While fixing cells captures them at a specific moment in a particular state (behavioral or structural), imaging live cells provides a comprehensive view of dynamic biological processes as they unfold. Live cell microscopy is a powerful technique for studying spatiotemporal changes in cells in real time. Through time-lapse video microscopy, researchers can track cells over extended periods, capturing dynamic processes such as structural reorganizations or transient events that would be undetectable in fixed-cell preparations. Additionally, with a microscope that can monitor cells inside an incubator, cells experience less environmental stress, leading to more reliable and consistent experimental data, because they remain in the incubator. Fluorescence microscopy of live cells is now also a crucial technique in contemporary cell biology. The use of fluorescent protein tags, live cell dyes, and other labeling methods enables the visualization of specific proteins, offering a variety of tools to study nearly any cellular process under the microscope.

3.0 Scope (Provide an overview of the PWS emphasizing the most important aspects of your requirement and defining the boundaries of the contractor’s performance responsibilities).

Research performed in the Wang Lab involves observation and analysis of cells from humans, mice, or rats, such as mesenchymal stem cells and pancreatic beta cells. For this goal, we require a piece of equipment that allows for real-time observation of multiple cell culture plates and dishes in an incubator. The JuLI™ Stage has a fully automated x-y-z stage and multi-channel fluorescent colors to acquire cell images and videos from various cell culture plates (up to 384wells) and dishes in an incubator. This piece of equipment will come with a stand-alone computer and software, and will not interface with the VA system. The system must meet the following minimum requirements:

· High-performance system with a wide operating temperature range from 5°C to 40°C

· Capacity to monitor multiple cell culture plates or dishes at once (up to up to 384 wells)

· MUST have small footprint of (W x D x H) 429 x 310 x 324 mm to fit in the designated laboratory incubator space

· MUST be compatible with Spheriod STAT Software (or equivalent) allowing recording and analysis of cell activity

· MUST support both Standard and Fast run modes

· MUST support brightfield and three fluorescence channels, three objective lenses, and sensitive filter-based optics to optimize for various live cell assays

· MUST have an automatic XY-stage to precisely navigate to the intended position as well as a holder that is

· manufactured after SBS standard allowing insertion of standard microplates

· MUST have interchangeable objective lens (4x, 10x, 20x) to allow imagery of cells at various magnification levels

· MUST have simple recalibration procedures to maintain optimal performance

· MUST have an organized, simple user interface for optimal analysis of data

· MUST be able to interface with a computer (PC compatible) or network (Ethernet LAN/WiFi-enabled) if needed

· MUST collect data for all channels and all wells regardless of plate setup, to allow for easy setup-error correction

· MUST be intuitive, and provide convenient & flexible system software

· MUST be suitable for the following analysis applications: Live cell imaging, Cell culture quality control, Cell culture optimization, Cell-based assay optimization, Angiogenesis, Protein expression, Kinetic imaging and cell differentiation, Kinetic imaging and cell differentiation, Stem cell development, Cell proliferation, Cytotoxicity, Cell migration.

4.0 Applicable Directives (List all documents referenced in the next section 5.0, Performance Requirements, with which the contractor will have to comply in performance of the contract effort. If only portions of a given document apply, specifically identify the applicable portions).

Not applicable. No contractors are required.

5.0 Performance Requirements (Identify what the contractor is required to do (major tasks and subtasks).

Not applicable. No contractors are required.

6.0 Deliverables (List all data required to manage or monitor contractor performance).

Not applicable. No contractors are required.
