STATEMENT OF WORK (SERVICES)
(SOW)
GENERAL INFORMATION
Title of Project:
Whole Proteome and Phospho Proteome Analysis on Brain Samples Statement of Need and Purpose:
The Stress and Addiction Neuroscience Unit (SANU) of the Integrative Neuroscience Research Branch (INRB) of The National Institute on Drug Abuse (NIDA) Intramural Research Program (IRP) was established for the purpose of understanding the neurobiology of substance use disorder-related and stress-related behaviors, with a particular focus on the contribution of the glucocorticoid system on these behaviors. The SANU is in need of whole proteome and phospho proteome analysis on brain samples. This service is required to measure protein, phosphorylated protein, and peptide levels in four addiction-related brain regions in opioid and alcohol dependent rodents. The results will provide a comprehensive understanding of the molecular changes that are produced by drug dependence and their modulation by glucocorticoid receptors.
Background Information and Objective:
The National Institutes of Health (NIH) is the nation’s leading medical research agency and the primary federal agency whose mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability, conducting, supporting and making medical discoveries that improve people’s health and save lives.
The National Institute on Drug Abuse (NIDA) Intramural Research Program (IRP) is part of the NIH which mission is to conduct state-of-the-art research on basic mechanisms that underlie substance use disorders (SUDs), and to develop new methods for the treatment of SUDs.
Research is supported at the molecular, genetic, cellular, animal, and human clinical levels and conceptually integrates highly innovative strategies and techniques. The long-term goal of the research is to better understand the biological and behavioral factors contributing to initiation, maintenance, and elimination of SUDs (and associated diseases), and to translate this knowledge into improved strategies for prevention, treatment, and reduction of negative consequences for the individual and for society caused by substance use disorders.
Opioid and alcohol use disorders are major public health issues. The overarching goal of the Stress and Addiction Neuroscience Unit of NIDA IRP is to understand the underlying neurobiolological mechanisms of these disorders in order to facilitate the development of new strategies for prevention, diagnosis, and treatment. By using highly valid preclinical models of opioid and alcohol use disorders, the present project aims to examine global changes in protein, phosphorylated proteins, and peptide levels in brain regions associated with the three domains in substance use disorder: binge intoxication (basal ganglia), withdrawal/negative affect
(extended amygdala), and executive function (cortices). This project will involve pharmacoproteomics to understand the role of glucocorticoid receptors, which are dysregulated in opioid and alcohol use disorder, on protein, phosphorylated proteins, and protein levels. This is a new project that is part of the program of research in NIDA IRP’s Stress and Addiction Neuroscience Unit (PI: Dr. Leandro F. Vendruscolo).

September 25, 2023 – September 24, 2024 (estimated)

SCOPE OF WORK
General Requirements:
Independently and not as an agent of the Government, the Contractor shall furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government as needed to perform the Statement of Work below:
The contractor is responsible for ensuring that the services are performed in a timely manner with a low error rate. The contractor shall provide all labor and material necessary to perform the specific requirements set forth below.
Specific Requirements:
The Contractor shall provide Whole Proteome and Phospho Proteome Analysis on Brain Samples during the 12-month period described in the General Requirements section, and to include at minimum the following:
1. Brain samples from opioid and and alcohol dependent rodents will be analyzed via mass spectrometry to determine proteome and phosphoproteome changes associated with alcohol and opioid dependence. Approximately 192 individual samples are to be analyzed.
2. Analysis of brain samples from alcohol and opioid dependent rats that were treated with a potential therapeutic for substance use disorders.
3. Analysis of brain samples shall include specific brain regions. The brain regions to be analyzed are all implicated in various components of substance use disorders and include the central nucleus of the amygdala, paraventricular nucleus of the hypothalamus, insula, and nucleus accumbens.
4. Analysis must include specific proteins and phosphoproteins. The proteins and phosphoproteins of interest within these brain regions include small peptides associated with stress and addiction that are historically difficult to measure with traditional protein analysis techniques, such as corticotropin-releasing factor, vasopressin, neuropeptide Y, hypocretin, oxytocin, dynorphin, and ghrelin. Other proteins of interest include the receptors of these peptides and proteins associated with stress signaling, gene expression, hormone synthesis, and circadian rhythm regulation.
5. Analysis must be a statistically valid comparison of 4 experimental groups (differential expression analysis of a 2x2 experimental design).
GOVERNMENT RESPONSIBILITIES
Following a psychopharmacological approach in rodents made dependent on opioids or alcohol, we will collect and provide several brain regions to be processed by the vendor.
DELIVERY OR DELIVERABLES
The deliverables shall be provided as electronic reports in the form of excel files. They will contain data analysis indicating the levels of expression of phosphoprotein and proteins of interest within each brain region analyzed from each individual animal. The deliverables will also include a bioinformatic differential expression analysis of the proteins across our treatment groups and figures illustrating these data (volcano plots and heatmaps). The files will be sent via email. Deliverables are to be received between 2023-2024.
REPORTING REQUIREMENTS
The contractor is not required to provide written progress reports, only the final report for each batch of processed samples as described in the deliverables.

Travel:
There will be no required contractor travel; no reimbursable expenses are expected.

Data Rights:
The contractor will produce testing results in a safe and secure manner. Contractor will have no rights to data.
The National Institute on Drug Abuse (NIDA) shall have unlimited rights to and ownership of all deliverables provided under this contract including reports, recommendations, briefings, work plans and all other deliverables. This includes the deliverables provided under the basic contract and any optional task deliverables exercised by the contracting officer. In addition, it includes any additional deliverables required by contract change. The definition of “unlimited rights” is contained in Federal Acquisition Regulation (FAR) 27.401, “Definitions.” FAR clause 52.227-14, “Rights in Data-General, ” is hereby incorporated by reference and made a part of this contract/order.
Section 508—Electronic and Information Technology Standards:
The contractor is not required to develop, procure, maintain, or use Electronic and Information Technology (EIT) in the performance of the SOW.
Publications and Publicity:
The contractor will not write and publish articles of data produced as a result of the requirements of the SOW.
Confidentiality of Information:
The Contractor may not disclose any information they obtain which is designated by NIDA as confidential or proprietary. The Contractor will use reasonable efforts to protect against any such disclosure. Testing results are expected to be considered confidential information.

The COR is responsible for: (1) monitoring the Contractor's technical progress, including the surveillance and assessment of performance and recommending to the Contracting Officer changes in requirements; (2) interpreting the Statement of Work and any other technical performance requirements; (3) performing technical evaluations as required; (4) performing technical inspections and acceptances required by this contract; and (5) assisting in the resolution of technical problems encountered during performance.
The Contracting Officer is the only person with authority to act as agent of the Government under this contract. Only the Contracting Officer has authority to: (1) direct or negotiate any changes in the statement of work; (2) modify or extend the period of performance; (3) change the delivery schedule; (4) authorize reimbursement to the Contractor for any costs incurred during the performance of this contract; (5) otherwise change any terms and conditions of this contract; or (6) sign written licensing agreements.
Any signed agreement shall be incorporated by reference in the contract.
The Government may unilaterally change the COR designation for this contract.

The following clause is applicable to all Purchase Orders, Task or Delivery Orders, and Blanket Purchase Agreement (BPA) Orders/Calls: Prompt Payment (Jan 2017) FAR 52.232-25. Highlights of this clause and NIH implementation requirements follow:
A. An invoice is the Contractor's bill or written request for payment under the contract for supplies delivered or services performed. A proper invoice is an "Original" which must include the items listed in subdivisions 1 through 12, below, in addition to the requirements of FAR 32.9. If the invoice does not comply with these requirements, the Contractor will be notified of the defect within 7 days after the date the designated billing office received the invoice (3 days for meat, meat food products, or fish, and 5 days for perishable agricultural commodities, dairy products, edible fats or oils) with a statement of the reasons why it is not a proper invoice. (See exceptions under II., below.) Untimely notification will be taken into account in the computation of any interest penalty owed the Contractor.
1. Contractor: Name, Address, Point of Contact for the invoice (Name, title, telephone number, e-mail and mailing address of point of contact).
2. Remit-to address (Name and complete mailing address to send payment).
3. Remittance name must match exactly with name on original order/contract. If the Remittance name differs from the Legal Business Name, then both names must appear on the invoice.
4. Invoice date.
5. Unique invoice #s for all invoices per contractor regardless of site.
6. NBS document number formats must be included for awards created in the NBS: Contract Number;
Purchase Order Number; Task or Delivery Order Number and Source Award Number (e.g., Indefinite Delivery Contract number; General Services Administration number); or, BPA Call Number and BPA Parent Award Number.
7. Unique Entity Identifier (UEI) as registered in the System for Award Management (SAM).
8. Federal Taxpayer Identification Number (TIN). In those exceptional cases where a contractor does not have a DUNS number or TIN, a Vendor Identification Number (VIN) must be referenced on the award document.
9. Identify that payment is to be made using a three-way match.
10. Description of supplies/services the description on the award, by line billed.*
11. Freight or delivery charge must be billed as shown on the award. If it is included in the item price do not bill it separately. If identified in the award as a separate line item, it must be billed separately.
12. Quantity, Unit of Measure, Unit Price, Extended Price of supplies delivered or services performed, as applicable, and that the line items specified in the award.*
* NOTE: If your invoice must differ from the line items on the award, please contact the Contracting Officer before submitting the invoice. A modification to the order or contract may be needed before the invoice can be submitted and paid.

Shipping costs will be reimbursed only if authorized by the Contract/Purchase Order. If authorized, shipping costs must be itemized. Where shipping costs exceed $100, the invoice must be supported by a bill of lading or a paid carrier's receipt.

A. Except as indicated in paragraph B., below, the due date for making invoice payments by the designated payment office shall be the later of the following two events:
1. The 30th day after the designated billing office has received a proper invoice.
2. The 30th day after Government acceptance of supplies delivered or services performed.
B. The due date for making invoice payments for meat and meat food products, perishable agricultural commodities, dairy products, and edible fats or oils, shall be in accordance with the Prompt Payment Act, as amended.
A. An interest penalty shall be paid automatically, if payment is not made by the due date and the conditions listed below are met, if applicable.
1. A proper invoice was received by the designated billing office.
2. A receiving report or other Government documentation authorizing payment was processed and

3. In the case of a final invoice for any balance of funds due the contractor for supplies delivered or services performed, the amount was not subject to further settlement actions between the Government and the Contractor.
B. Determination of interest and penalties due will be made in accordance with the provisions of the Prompt Payment Act, as amended, the Contract Disputes Act, and regulations issued by the Office of Management and Budget.
Upon receipt of accelerated payments from the Government, the Contractor shall make accelerated payments to its small business subcontractors under this contract, to the maximum extent practicable and prior to when such payment is otherwise required under the applicable contract or subcontract, after receipt of a proper invoice and all other required documentation from the small business subcontractor.
The acceleration of payments under this clause does not provide any new rights under the prompt Payment Act.
Include the substance of this clause, include this paragraph c, in all subcontracts with small business concerns, including subcontracts with small business concerns for the acquisition of commercial items.
Definitions. As used in this clause - Payment request means a bill, voucher, invoice, or request for contract financing payment with associated supporting documentation. The payment request must comply with the requirements identified in FAR 32.905(b), "Content of Invoices" and the applicable Payment clause included in this contract.
Except as provided in paragraph (c) of this clause, the Contractor shall submit payment requests electronically using the Department of Treasury Invoice Processing Platform (IPP) or successor system. Information regarding IPP, including IPP Customer Support contact information, is available at www.ipp.gov or any successor site.
The Contractor may submit payment requests using other than IPP only when the Contracting Officer authorizes alternate procedures in writing in accordance with HHS procedures.
If alternate payment procedures are authorized, the Contractor shall include a copy of the Contracting Officer's written authorization with each payment request.
