420 and Ztea View PMX-120
Solicitation No.: 75N91022Q00135 Posted Date: 07/26/2022 Response Date: 08/03/2022 by 12:00 PM EST Classification Code: 6640 (Laboratory Equipment and Supply) NAICS Code: 334516 (Analytical Instrument Manufacturing)
The Department of Health and Human Services (HHS), National Institutes of Health (NIH), National Cancer Institute (NCI), Center for Cancer Research (CCR), Neuro-Oncology Branch (NOB) and Laboratory of Pathology (LP) have a need for Brand Name or Equal Particle Metrix Inc. ZetaView QUATT PMX-420 and Ztea View PMX-120.
This is a Combined Synopsis/Solicitation for commercial items, prepared in accordance with the format in Federal Acquisition Regulation (FAR) subpart 12.6, as supplemented with additional information included in this notice, and shall be processed in accordance with FAR Part 13.106 – Simplified Acquisition Procedures. This announcement constitutes the only solicitation that will be provided in relation to the stated need contained herein. This solicitation, 75N91022Q00135, is issued as a request for quote (RFQ) and includes applicable provisions and clauses in effect through FAR Federal Acquisition Circular (FAC) 2022-06 (05/26/2022).
This requirement is a Total Small Business Set-Aside under the North American Industry Classification Code (NAICS) 334516 (Analytical Laboratory Instrument Manufacturing) for which the business size standard 1,000 employees. One Firm-Fixed-Priced award is anticipated to be made in relation to this solicitation.
1.0 BACKGROUND
The Department of Health and Human Services (DHHS), National Institutes of Health (NIH), National Cancer Institute (NCI), Center for Cancer Research (CCR) has many laboratories engaged in research efforts associated with cancer. Two of these laboratories are the Neuro-Oncology Branch (NOB) and Laboratory of Pathology (LP). Both laboratories currently require Multiple Parameter Particle Tracking Analyzers in support of their ongoing research efforts.
The Neuro-Oncology Branch (NOB) is performing translational research on central nerve system (CNS) tumors–\ using biological, biochemical, pathological, and immunological methods to identify underlying mechanisms of patient-reported outcomes and develop novel therapeutics for the patients with brain tumors. NOB is investigating the biologic underpinnings of critical symptoms with the goal to improve symptom management and patient outcomes. A Multiple Parameter Nanoparticle Tracking Analyzer (MPNTA) instrument that allows fast and reliable measurement of size, concentration, and Zeta Potential of nanoparticles, such as extracellular vesicle (EV). In contrast to scatter-based particle detection, multi-fluorescence MPNTA the next generation EV characterization fluorescence detection capability of instrument allows the user to get biochemical information about particles. This increases the resolution and discriminatory power of the data.
The Laboratory of Pathology (LP), Translational Nanobiology Section requires the purchase of a Multiple Parameter Nanoparticle Tracking Analyzer to replace a legacy Nanosight LM-10 instrument that is no longer manufactured or fully supported / repairable.
2.0 BRAND NAME OR EQUAL
This requirement is for the procurement of the brand name or equivalent instruments described in Section
4.1. The Federal Acquisition Regulation (FAR) Provision FAR 52.211-6, Brand Name or Equal (AUG 1999) is applicable to this requirement.
3.0 TYPE OF ORDER
This is a Firm Fixed-Price Purchase Order.
4.0 SPECIAL ORDER REQUIREMENTS
4.1 PRODUCT FEATURES/SALIENT CHARACTERISTICS
4.1.1 One (1) Brand Name or Equal ZetaView QUATT PMX -420
The following product features/characteristics are required for this requirement:
Automated Particle Tracking Analyzer with 90° Multi-Source Scattering Video Microscope for the determination of particle size distribution, particle concentration, zeta potential distribution, particle mobility and fluorescence detection mode.
Must be equipped with Cell cassette and sample cell, 10x microscope objective, metal-oxide semiconductor (CMOS) camera, Instrument Controller with software installed, ready for operation
Must have slide-in cell assembly, Peltier temperature control from 5°C below RT up to 45°C, with a minimum of three (3) fluidics channels: two for rinsing, one for volume transport of wastes.
Must be able to recognize and distinguish whether the particles are biological vesicles, protein aggregates, inorganic precipitates or simply cell waste.
Must have the fluorescence detection capability to get biochemical information about particles, thus increasing the resolution and discriminatory power of the data.
Must have the measurement technology and present the latest fluorescent nanoparticle tracking analysis (f-NTA) results on extracellular vesicle phenotyping using membrane dyes and monoclonal antibodies.
Must have a minimum four LASER sources and filters, pre-aligned and computer selectable; No manual exchange of LASERS, which then would require a new sample and excessive time to exchange.
Must provide 405/488/520/640 combination with appropriate long pass filters for fluorescence measurements. Automated 5-position filter wheel with 4 Long Wave Pass (LWP) emission filters, cut-off @ 430 nm & 500 nm 550 nm & 680 nm
Must be capable of zeta potential measurements on the same sample volume used to measure size, concentration and fluorescence.
Instrument must be able to collect data across multiple sub-volume positions within the sample chamber by use of automated camera-lens-laser positioning used for sizing, concentration, zeta potential and fluorescence measurements. In doing so, the sampling statistics of tracking nanoscale volumes are greatly improved, relative to the fixed-position-sampling.
Sample cell walls must consist of chemically inert fused silica, and support use of multiple sub-volume sampling across more than 500 microns of range, in order to provide for proper sampling efficiency.
Sample cell must be easy to clean and be chemically robust, and large enough to work well with the integrated multi-point sampling.
Instrument must use minimum 10x objective lens or better, for up to 40x increased sampling volume. As per multiple sub-volume sampling, sampling a larger volume of suspension in each sub-volume further improves NTA statistics. Syringe pumps are not adequate due to their negative influence on size and concentration measurements.
Instrument software must provide real-time particle counting of the live video output, whereby the # particles/frame is determined BEFORE measurement and analysis. Having live, real time tracking ability allows one to optimize data collection and analysis, to better build robust SOPs.
Otherwise, poor-quality data could result.
Instrument software must provide a digital CMOS projection of light scattering particles within the real-time live video output, in addition to standard analog video CMOS presentation. Digital projection of tracking efficiency, real time, allows for measurement optimization
Must provide simultaneously aligned, software-controlled 4 sources for fluorescence NTA measurements. The Controller Software must be able to determine, particle size & concentration, zeta potential, and cell temperature. Flush pump & temperature control.
Auto-alignment & auto-focus. Incremental sub-volume sampling through cell. Daily instrument performance check. Automated filter selection for all source wavelengths with F-NTA detection.
Full data reporting via PDF and TXT files.
One second videos must be statistically practical, allowing for F-NTA data collection before photo bleaching occurs for most fluorophores.
Linear concentration range must be at least 5e6 to 3e8 particles per ml.
Automated fast scanning multi-point sampling. Instrument must allow fast and reliable measurement of size, concentration, and Zeta Potential of nanoparticles in the size range from 10 nm to 2 µm and for a statistically valid data set within two minutes.
4.1.2 One (1) Brand Name or Equal ZetaView PMX-120
The following product features/characteristics are required for this requirement:
Measure concentration and size of nanoscale/submicron particles.
Be configured with a fixed, 488nm, single laser system, capable of being upgraded to a minimum of three lasers.
Small, recoverable sample volume of ≤ 600µL.
Minimum of 0.5E6 concentration sensitivity.
