5. SCOPE OF WORK:

Environmental chemistry method for soil and independent laboratory validation (OCSPP 850.6100) for 3-chloro-4-methylaniline hydrochloride (CAS No. 7745-89-3)

5.1 BACKGROUND

As part of registration review of the pesticide active ingredient 3-chloro-4-methylaniline hydrochloride (CAS No. 7745-89-3), the Environmental Protection Agency (EPA) is requiring submittal of an environmental chemistry method (ECM) and independent laboratory validation (ILV) for soil with a maximum method limit of quantification (MLOQ) of 15 µg active ingredient per kg of soil. The ECM(s) and ILV must be conducted in accordance with EPA’s OCSPP 850.6100: Environmental Chemistry Methods and Associated Independent Laboratory Validation [1], 40 CFR Part 136 Appendix B [2], Federal Insecticide, Fungicide, and Rodenticide Act (FIFRA) Good Laboratory Practice (GLP) requirements [3], and EPA PRN 2011-3: Standard Format for Data Submitted Under FIFRA and Certain Provisions of FFDCA [4].

12639522Q0186 Solicitation for Soil Analysis for USDA APHIS NWRC Ft Collins, CO 3 of 39 http://www.sam.gov/ http://www.aptac-us.org/new/Govt_Contracting/find.php mailto:thomas.j.wesley@usda.gov

5.2 OBJECTIVES

The objectives are:

1) Develop, write, and validate an ECM for one soil matrix for two analytes: the pesticide active ingredient 3-chloro-4-methylaniline hydrochloride (CAS No. 7745-89-3) and its primary metabolite in soil, 3-chloro-4-methyl acetanilide (CAS No. 7149-79-3). The ECM must have a maximum MLOQ of 15 µg active ingredient per kg of soil. No specific MLOQ is required for the metabolite.

2) Complete an ILV of the ECM in objective 1.

5.3 TASKS OR REQUIREMENTS

The Contractor is required to provide all labor and materials required to complete the following deliverables (unless otherwise specified), but not in this order:

• Draft GLP study protocols for the validation of the ECM and ILV. Study protocols must be approved by the study sponsor's representative (USDA Technical Contact) prior to initiation of any laboratory testing. Any amendments to the study protocols will also be approved by the sponsor's representative prior to implementation.

• Requirements for the ILV: The ILV study provided by or subcontracted by the Contractor must be completed by a different laboratory and study director and use different analysts, equipment, instruments, and supplies than the laboratory that develops and validates the ECM as described in OPPTS 850.6100 [1]. The laboratories or facilities conducting the ECM and ILV must be distinct and operated separately and without collusion. The analysts and study director of the ILV must be unfamiliar with the ECM development or validation prior to the ILV

• After consultation with the sponsor’s representative (USDA Technical Contact), procure the soil matrix from a U.S. source, which is GLP-characterized for organic carbon content, clay content, soil texture, pH, cation exchange capacity, and specific surface for both the ECM and ILV studies.

• Procure and verify the purity of reference standards for 3-chloro-4-methylaniline hydrochloride (CAS No. 7745-89-3) and 3-chloro-4-methyl acetanilide (CAS No. 7149- 79-3). The test substance, 3-chloro-4-methylaniline hydrochloride, will be provided to the contract laboratory by the USDA Technical Contact already GLP-characterized.

• Develop an ECM for soil that use Gas Chromatography/Mass Spectrometry (GC/MS) or Liquid Chromatography/Mass Spectrometry (LC/MS), or additionally provide a confirmatory technique as required in OPPTS 850.6100 [1]. The ECM’s mean recoveries for a minimum of 5 spiked samples at the LOQ and at 10 × LOQ must be within 70- 120% and the relative standard deviation ≤20% for the soil matrix, or justification for why these performance criteria cannot be achieved must be provided.

• Conduct the validation of the ECM and ILV in accordance with the requirements listed in OCSPP 850.6100 [1], 40 CFR Part 136 Appendix B [2], and FIFRA GLPs [3]. Up to 3 attempts may be required for the ILV as allowed in OPPTS 850.6100 [1].

• Draft GLP final study reports for the validation of the ECM and ILV. Final reports must include representative chromatograms for each fortification level, controls, reagent blanks, and calibration standards, and verification that the ECM laboratory is distinct from the ILV laboratory, and other reporting requirements listed in OPPTS 850.6100 [1].

• Requirement for the ILV: Any contact with the registrant or developers during the establishment of the method should be documented in writing in the final report.

12639522Q0186 Solicitation for Soil Analysis for USDA APHIS NWRC Ft Collins, CO 4 of 39

• Obtain the review and approval for the ECM and ILV final reports from the sponsor’s representative (USDA Technical Contact). Once approved, provide signed final reports that meet the FIFRA GLP requirements [3] and EPA's data submission format requirements in EPA PRN 2011-3 [4].

• Disposal of the unused or no longer needed test substances and samples.

5.4 APPLICABLE STANDARDS

The Contractor must adhere to the standards as outlined below:

• All aspects of the study and study records will be completed in compliance with EPA OCSPP 850.6100 [1] except when agreed upon in writing ahead of time by the sponsor’s representative.

• All aspects of the studies and studies’ records will be completed in compliance with EPA FIFRA GLP standards.

• All deviations from EPA guidelines require written approval of the sponsor’s representative.

• Written approval of the sponsor’s representative may be obtained via e-mail, or signed study protocol or protocol amendment, as appropriate.

• If, during the EPA science review after submittal, EPA determines the validation of the ECM or ILV does not fulfill the data requirement because of the test facility’s errors or omissions in the final reports, or failure to adhere to the EPA study guideline and/or GLP standards during the study phase in ways that were not previously agreed upon by the sponsor’s representative, the vendor shall repeat the study at the vendor’s cost or refund the amount of the contract. If EPA does not find either study acceptable to fulfill the core data requirement for reasons unrelated to the test facility’s compliance with EPA guidance of regulations, the vendor is not responsible for failure to meet the terms of the contract.

• The final reports will be formatted to conform to EPA’s PRN 2011-3 [4].

• Any deviations from the EPA study guidelines must be well documented with sound justification in the final reports.

5.5 CONTRACTOR QUALIFICATIONS

The contractor shall have demonstrated successful experience in developing and validating environmental chemistry methods for soils and conducting ILVs in accordance with EPA FIFRA GLPs [3] as described in Section 3 below.

5.6 PLACE OF PERFORMANCE

The order shall be conducted within the Contractor’s facilities and any subcontracted company’s facilities.

5.7 PERIOD OF PERFORMANCE

The signed GLP final report shall be completed within 12 months after the Contractor receives the test substance from the Sponsor’s representative (USDA Technical Contact).

1.8 CONTRACT TYPE

The Government intends to issue this requirement as a firm fixed price purchase order.
