About this file

This Performance Work Statement (PWS) outlines requirements for cyanotoxin testing and material procurement for the Department of Interior U.S. Geological Survey (USGS) National Wildlife Health Center (NWHC) in Madison, WI. The contractor must provide analytical services to detect cyanotoxins in various tissue specimens from birds, mammals, amphibians, and reptiles using ELISA and/or LC-MS/MS testing methods. Specific testing capabilities must include detection of Microcystins/Nodularins, Cylindrospermopsin, Anatoxin-a, Saxitoxins, Dermatoxins, Domoic Acid, Brevetoxin, Okadaic Acid, and BMAA.

The contractor must also provide algal material for dosing experiments, including capabilities to lyophilize natural algal biomass, characterize it for cyanotoxins, provide material in gelatin caplets, and meet specified toxin concentrations. Quality control requirements include laboratory duplicates within 20% coefficient of variation and specific quality assurance parameters for spiked samples. Deliverables include test results within 90 days via email, detailed documentation of methods and quality control, and Certificates of Analysis for purchased algal material. The contractor is responsible for implementing quality control measures before returning results to the Government.

PERFORMANCE-BASED WORK STATEMENT (PWS)

Cyanotoxin testing and purchase of cyanotoxin material.

Background.

The Department of the Interior (DOI) U.S. Geological Survey (USGS) National Wildlife Health Center (NWHC) investigates the effects of cyanotoxin exposure in wildlife experimentally and from wildlife morbidity and mortality events.

The NWHC requires high quality cyanotoxin testing of tissues and other matrixes and purchase of cyanotoxin dosing material for experimental studies.

SCOPE: Services, non-personal, to provide all plant equipment, labor, travel, and materials (unless otherwise provided herein) necessary for processing of National Wildlife Health Center – Madison, WI, (NWHC) cyanotoxin tissue specimens for presence of cyanotoxins for birds, mammals, amphibians, and reptiles. Services, non-personal, to provide all plant equipment, labor, travel, and materials (unless otherwise provided herein) necessary for processing natural algal material for use in dosing experiments.

REQUIREMENTS:

The Contractor shall provide:

I. Analytical services to detect cyantoxins.

1.0 Routine testing by ELISA and/or LC-MS/MS for cyanotoxins. The contractor shall have the ability to:

1.0.1 Process a wide variety of tissue types from a wide variety of taxa.

1.0.2 Test for Microcystins/Nodularins, Cylindrospermopsin, Anatoxin-a, Saxitoxins, Dermatoxins, Domoic Acid, Brevetoxin, Okadaic Acid, and BMAA.

1.0.3 Perform MMPB testing for Microcystins/Nodularins

1.0.4 Perform LC-MS/MS for a suite of Microcystins (14+ variants)

1.0.5 Perform ELISA for Microcystins/Nodularins, Saxitoxins, and Brevetoxins.

1.0.6 Perform LC-MS/MS for Microsystins/Nodularins, Cylindrospermopsin, Anatoxin-a, Saxitoxins, Dermatoxins, Domoic Acid, Brevetoixns, Okadaic Acid, and BMAA.

1.0.7 Perform quality control testing in association with specific toxin analysis.

1.0.8 Laboratory duplicates and laboratory fortified matrix duplicates shall be within 20% coefficient of variation. Quality assurance/quality control samples shall have a %return of +/-50% or better when the spiked concentration of the laboratory fortified sample matrix when fortified concentration is at 2 x minimum detection limit, the isotope shall have a %return of +/- 50% or better, and the laboratory fortified sample matrix shall have a %Return of +/- 30% of spiked concentration.

II. The Contractor shall provide:

1.0 Algal material to be used in dosing experiments. The contractor shall have the ability to:

1.0.1 Lyophilize natural algal biomass.

1.0.2 Characterize natural algal biomass for cyanotoxins.

1.0.3 Provide lyophilized material in gelatin caplets.

1.0.4 Provide material to the toxin specifications provided by the Contracting Officer's

Technical Representative (COTR) or their designee with regards to toxin concentration.

DELIVERABLES:

1. All test results produced shall be delivered to the Contracting Officer's Technical Representative (COTR) or their designee in accordance with the instructions provided in the contract. Results to be delivered by email within 90 days of submission.

2. Results should include detailed description of sample preparation, including tissue hongenization and extraction methods, analytical methods, quality control results, and results of tissue or other matrix testing. Results should include interpretation of any abbreviations or acronyms used.

3. For purchased algal material, a Certificate of Analysis Report will accompany the material itself detailing the material provided, analysis conducted to evaluate the material for toxin presence and concentration, methods of sample preparation/extraction, quality control results, and analysis results.
