About this file
This scope of work document outlines a project to estimate the ages of 30 humpback whales through amino acid racemization analysis of eye lens tissue. Key aspects of the project include dissecting and preparing whale eyeball samples from Alaska and Hawaii for chemical analysis by November 2023. The contractor will then conduct amino acid racemization analyses on the samples by December 2023 and provide a report on findings to the National Park Service by January 2024. The project aims to estimate whale ages through comparison to a baseline ratio of aspartic acid isomers in calf eyeballs. Additional details may be provided by the National Park Service based on known ages or anatomical data of sampled whales. The related federal contract opportunity is solicitation number 140P9723Q0080 from the Department of the Interior National Park Service Alaska Region to conduct amino acid racemization age estimates on Alaska humpback whale eye lens tissues.

SCOPE OF WORK
Alaska Humpback Whale AAR Age Estimates
PR-GLBA-A-039
This project aims to perform amino acid racemization (AAR) chemical analysis on whale eye lens tissue from 30 humpback whale eyes to estimate each whale's age and produce a report containing the results and interpretation of the findings. Seven eyes have been dissected, and the lens nucleus tissue is provided. Twenty-two are whole eyes that need to be dissected. One extra is included.
Overall, four components will make up this project in collaboration, including:
(1) Receipt of the frozen whale eyeballs from Alaska and dissecting the eye lens nucleus using previously published techniques.
(2) Receipt of frozen whale calf eyeballs from Hawaii and dissecting the eye lens nucleus using previously published techniques.
(3) Amino acid racemization analyses on each of the calf and non-calf eyeballs. The calf eyeballs provide a baseline ratio of D/L isomers of aspartic acid that forms the basis of comparison for all the other whale eye lenses and allows age determination.
(4) A consultation will be done by the NPS with GLBA whale biologists on the known age, independently estimate age, age class, or anatomical details of whales that may assist in the
AAR estimate of each whale’s age and provided to the contractor as ancillary information on age
(5) Compiling the results into a report to be sent to GLBA and shared with other collaborators on the project.
Timeline: The frozen whale tissue will be shipped to the contractor. The laboratory will dissect and prepare the tissues for chemical analysis by November 2023, conduct the AAR analysis by December
2023, and provide a report on their findings by January 2024.
After completion of the services purchased here, it is anticipated that the contractor will work closely with NPS and other collaborators to help develop a manuscript to be submitted for publication in peer-reviewed scientific journals.
