PERFORMANCE WORK STATEMENT
PERFORMANCE WORK STATEMENT (PWS)
Isolation and Purification of Ovatoxin-a from O. Ovata Cell Pelle United States Army Medical Research Institute of Infectious Diseases (USAMRIID)
SECTION 1 PURPOSE
1.1. Introduction
The United States Army Medical Research Institute of Infectious Diseases (USARMIID) is located in Fort Detrick, MD. Since 1969, USAMRIID has served as the Department of Defense’s (DoD) lead laboratory for medical biological defense research. Research conducted at USAMRIID leads to medical solutions – therapeutics, vaccines, diagnostics, and information that benefit both military personnel and civilians. USAMRIID is a subordinate laboratory of the U.S. Army Medical Research and Development Command (MRDC). The Diagnostic Systems Division within USAMRIID is responsible for developing an animal model of palytoxin (PLTX) aerosol toxicology for testing of potential medical countermeasures. This effort requires the development of good diagnostic methods to support aerosol toxin exposure. As with all good diagnostics, information on how the methods perform on toxins with similar structure is critical.
1.2. Mission
Provide leading edge medical capabilities to deter and defend against current and emerging biological threat agents.
1.3. Background
USAMRIID is requesting a service contract to isolate and purify ovatoxin-a from O. ovata cell pellets provided by USAMRIID. This would allow USAMRIID to acquire sufficient purified toxins to support comparative aerosol toxicology studies and diagnostic method development.
1.4. Scope
1.4.1. The Contractor shall optimize and further develop their existing protocol for isolation of ovatoxins
1.4.2. The Contractor shall isolate and purify ovatoxin-a from O. ovata cell pellets provided by the USAMRIID.
1.4.3. Once complete, the contractor shall coordinate transfer of viable samples to the USAMRIID.
1.4.3.1 The Contractor shall ship/manage, store, and coordinate the shipment of investigational product(s) and study-related records in accordance with applicable regulation and laws, both foreign and domestic.
1.4.3.2 Ensure cold chain management and accountability of all investigational products used in relation to the study.
1.4.3.3 Purchase suitable shipping materials in adequate quantities to ensure successful product/record shipment.
1.4.3.3 Create and/or validate, or oversee the validation of, shipping procedures and methods.
1.4.3.4 Arrange for and/or provide a Broker(s) for international shipments (import and export), as necessary.
1.4.3.5 The Contractor shall provide the government a purification protocol/report supporting the service provided in this contract.

The contractor shall provide an OCI certification with their proposal. If there is an OCI, the contractor shall provide a draft OCI Mitigation Plan with their proposal.
1.5.2. The United States Army Medical Research and Development Command (USAMRDC) has categorized all service procurements into one of the following three categories as defined below, for the purpose of identifying, avoiding or mitigating Organizational Conflicts of Interest (OCI) in accordance with (IAW) Federal Acquisition Regulation (FAR) Subpart 9.5. These categories are defined as follows:
Category 1: Command Wide Internal Support: Services which, by their very nature, give the Contractor access to non-public procurement sensitive data about the contract requirements, other contractors’ proprietary information, or put the contractor in a position to influence the award decision on a contract in which they are participating. Services would be characterized as having access to Command Wide procurement sensitive information, or work that could likely have an influence over follow-on awards or contracts with subordinate directorates/Commands.
Category 2: Program Management Support: Services which assist USAMRMC in planning and managing an organization’s activities and programs. These include, for example: requirements analysis, acquisition support, budget planning and management, business process reengineering, program planning and execution support, and independent technical management support. Services would be characterized as having access to only Directorate or Subordinate Command level procurement sensitive information or work that would have little influence on follow-on requirements or requirements of other directorates/Subordinate Commands.
Category 3: Product Support. Services or end items required to meet the mission requirements of the organization’s activities and programs. These include, for example: concept exploration and development, system design, system development and integration, COTS procurement and integration, internal development testing, deployment, installation, operations, and maintenance. Services can be characterized as having little or no access to procurement sensitive information other than the immediate requirement and little chances of follow-on work that can be influenced by the immediate requirement.
OCI Certification. USAMRDC OCI Category 3 is applicable for this acquisition. The Contractor shall perform a thorough comparative analysis of the potential new work against all current and previous works performed in support of the USAMRIID organization and ascertain whether they have a possible conflict of interest. If there is a conflict of interest or the appearance of a conflict of interest, the Contractor shall provide a comprehensive OCI Mitigation Plan. The Contractor shall complete the OCI Certification at time of proposal submission (Deliverable 6, Exhibit 1).

This QASP is a “living document” and the Government may review and revise it on a regular basis. However, the Government shall coordinate changes with the contractor. Updates shall ensure that the QASP remains a valid, useful, and enforceable document. Copies of the original QASP and revisions shall be provided to the contractor and Government officials implementing surveillance activities.

4. PERFORMANCE STANDARDS.
Performance standards define desired services. The Government performs surveillance to determine if the contractor exceeds, meets or does not meet these standards.
The Performance Requirements Summary Matrix, shown below, includes performance standards. The Government shall use these standards to determine contractor performance and shall compare contractor performance to the Acceptable Quality Level (AQL).
PWS Reference Deliverable Indicator Standard Acceptable Quality Level Method of Surveillance PRS # 1 A list of samples (Isolated and Purified ovatoxin-a from O. ovata cell pellets) and a brief report on sample condition upon arrival will be sent to USAMRIID> 90% in purity invitro assay confirmed> 90% in purity invitro assay confirmed Validated invitro Assay with confirmation of results PRS # 2 Purification protocol and report Purification protocol and report PRS # 3 Extraction of cell pellets Sample aliquots will be extracted with MeOH/water (1:1) and individual extracts combined to obtain a single homogenous extract. LC-HRMS will be performed to identify toxin profile and content. The extract will then be divided into aliquots and concentrated under nitrogen flow prior to lyophilization.
Deviation with COR concurrence Quantification of toxin content in the initial and concentrated extracts will be performed using the commercially-available Waco PLTX as the standard.
PRS # 4 Toxin Purification Purification will progress through a 3-step process involving Combiflash chromatography, and semi-preparative and analytical HPLC chromatography steps. At each step, fractions will be analyzed and all ovatoxin a-containing fractions combined and separated from fractions containing other ovatoxins.
Toxin profiles and quantity will be monitored by LC-MS. At the end of the third step, toxin purity and the presence of degradation products will be assessed FIA-HRMS and LC-HRMS. If the purity at this point does not measure at least 90%, a final concentration and analytical HPLC step will be performed.Validated invitro Assay with confirmation of results
5. INCENTIVES.
This contract is Firm-Fixed Price (FFP). No incentives are provided under this contract.
6. METHODS OF QA SURVEILLANCE.
Various methods exist to monitor performance. The COR shall use the surveillance methods listed below in the administration of this QASP.
Regardless of the surveillance method, the COR shall always contact the contractor's task manager or on-site representative when a defect is identified and inform the manager of the specifics of the problem. The COR shall be responsible for monitoring the contractor’s performance in meeting a specific performance standard/AQL.
Surveillance results may be used as the basis for actions (to include payment deductions) against the contractor. In such cases, the Inspection of Services clause in the Contract becomes the basis for the KO’s actions.
7. RATINGS.
Metrics and methods are designed to determine if performance exceeds, meets, or does not meet a given standard and acceptable quality level. A rating scale shall be used to determine a positive, neutral, or negative outcome. The following ratings shall be used:
EXCEPTIONAL: Performance significantly exceeds contract requirements to the Government’s benefit.
SATISFACTORY: Performance meets contractual requirements.
UNSATISFACTORY: Performance does not meet contractual requirements.
8. DOCUMENTING PERFORMANCE.
a. ACCEPTABLE PERFORMANCE.
The Government shall document positive performance. A report template is attached. Any report may become a part of the supporting documentation for fixed fee payments, award fee payments, or other actions.
b. UNACCEPTABLE PERFORMANCE.
When unacceptable performance occurs, the COR shall inform the contractor. This will normally be in writing unless circumstances necessitate verbal communication. In any case the COR shall document the discussion and place it in the COR file.
When the COR determines formal written communication is required, the COR shall prepare a Contract Discrepancy Report (CDR), and present it to the contractor's task manager or on-site representative. A CDR template is attached to this QASP.
The contractor shall acknowledge receipt of the CDR in writing. The CDR will specify if the contractor is required to prepare a corrective action plan to document how the contractor shall correct the unacceptable performance and avoid a recurrence. The CDR will also state how long after receipt the contractor has to present this corrective action plan to the COR. The Government shall review the contractor's corrective action plan to determine acceptability.
Any CDRs may become a part of the supporting documentation for contract payment deductions, fixed fee deductions, award fee nonpayment, or other actions deemed necessary by the KO.
9. FREQUENCY OF MEASUREMENT.
a. Frequency of Measurement.
During contract/order performance, the COR shall take periodic measurements, as specified in the AQL column of the Performance Standards Summary Matrix, and shall analyze whether the negotiated frequency of measurement is appropriate for the work being performed.
b. Frequency of Performance Assessment Meetings.
The COR shall meet with the contractor, as needed, to assess performance and shall provide a written assessment.
