About this file
This is a Statement of Need (SON) for the acquisition of a MiSeq i100 Plus Sequencing System for the Center for Cancer Research Genomics Core at the National Cancer Institute. The acquisition will be made as a Firm Fixed-Price Purchase Order and requires delivery and installation within four to twelve weeks after contract award at the National Cancer Institute facility located at 37 Convent Drive, MSC 4256, Bethesda, MD 20892-4256, with Liz Conner serving as the primary point of contact.
The MiSeq i100 Plus system must deliver up to 100 million reads (30 gigabases) per run with run times of approximately four hours to enable same-day results. Key technical requirements include sequencing chemistries compatible with other government-owned Illumina platforms, use of XLEAP-SBS chemistry for improved data accuracy and reliability, an integrated onboard DRAGEN platform for secondary data analysis, and application support for targeted sequencing panels and whole-genome sequencing. The contractor shall provide comprehensive training on equipment operation, software use, and associated workflows for National Cancer Institute researchers, conducted by a premium support scientist or qualified Illumina personnel within one week of installation, with additional training available on an as-needed basis. This acquisition consolidates existing aging instruments (Illumina iSeq and legacy MiSeq) into a single modern platform to reduce operational complexity, decrease consumables management requirements, improve run reliability, and lower total cost of ownership for the research center.

STATEMENT OF NEED (SON)
1.0 TITLE
MiSeq i100 Plus Sequencing System
2.0 BACKGROUND
The purpose of this acquisition is to replace the current Illumina iSeq and legacy MiSeq instruments with a single Illumina MiSeq i100 Series system to ensure reliable and sustainable sequencing services for the Center for Cancer Research Genomics Core. The existing instruments are aging, increasingly prone to downtime, and require separate service agreements and distinct consumables, which increases operational complexity, cost, and the risk of service interruptions.
Consolidating sequencing capabilities onto one modern platform will reduce the number of service contracts and significantly decrease the range of reagents and consumables that must be stocked and managed. Procuring this system as a shared Core resource provides Center for Cancer Research investigators with access to a current, fully supported sequencing platform and eliminates the need for individual laboratories or branches to purchase and maintain their own instruments, many of which are now obsolete.
The MiSeq i100 Series offers improved run reliability, streamlined workflows, and flexible support for small- and mid-scale sequencing projects, reducing reruns, staff time, and turnaround delays. This upgrade lowers total cost of ownership, improves operational efficiency, and supports cost-effective, centralized use of sequencing infrastructure across the Center for Cancer Research.
3.0 TYPE OF ORDER
This acquisition will be made as a Firm Fixed-Price Purchase Order.
