PURCHASE DESCRIPTION
1. Statement of Need and Purpose: This is for the purchase of 3D liver microtissues composed of human primary hepatocytes and a mixture of human primary hepatocyte, liver endothelial cells and kupffer cells. These 3D liver microtissues are prodcued by InSphero in 96- or 384-well plates, at one microtissue per well, and are shipped life for ready testing upon receving. These 3D liver microtissues will be used to study viral infectivity, effects of viral infectivity on liver fucntion biomarkers, and effect of anti-viral compounds on viral infection. The liver microtissues of different cell type composition will allow use to assess the role of the different cell types on the readouts above. If sucessful, this assay platform of 3D liver microtussues might be integrated in a panel of tissues to study panels of viruses and anti-infective drugs.
2. Background Information and Objective: There is a need to establish in vitro predicitve assays for drug discovery and development. The NCATS 3D Tissue Bioprinting Laboratory is assembling a portofilio of 3D tissue models in microplate format that can be used for drug testing. In order to accelerate the creation of a ready to use tissue platforms to be able to rapidly respond to health care crisis, like the COVID-19 pandemic, the 3D Tissue Bioprinting laboratory develops its own tissue models, but also looks for opportunities with 3D tissue models that are commercially available, well validated and ready to use. InSphero was indentif ied by scienitsts at the 3DTBL as a company that produces live fucntional 3D liver microtissues, in a ready to use HTS compatible multiwell plate format. We will use these 3D liver microtissues composed of human primary hepatocytes and a mixture of human primary hepatocyte, liver endothelial cells and kupffer cells to to study viral infectivity, effects of viral infectivity on liver fucntion biomarkers, and effect of anti-viral compounds on viral infection.
3. Generic Name of Product: Life and functional pre-assembled 3D liver tissues containing human primary hepatocytes (single cell type) and containing human primary hepatocyte, liver endothelial cells and kupffer cells, in a multiwell plate format (96- or 384-well plate), shipped life and ready to use.
4. Purchase Description: 3D InSight™ Human Liver Microtissues (multi-donor hepatocytes), one tissue in each well of a 384-well plate, 3D InSight™ Human Liver MT (multi-donor hepatocytes, co-culture with Kupffer cells and LECs), one tissue in each well of a 96-well plate, and media necessary to culture the liver tissues.
