Statement of Work

1.0 Background

California recently faced a multi‐year drought period and occurrence of similar events in the near future cannot be ruled out. From environmental and agricultural perspective it is important to breed drought tolerant cultivars with better water use efficiency which will also help support water conservation. Continuous availability of irrigation water is a major concern for lettuce growers in California who face significant economic losses due to diminishing water supplies. In this project USDA ARS will conduct a comprehensive metabolome analysis on lettuce varieties under drought and control conditions to further understand the role of the accumulated metabolites and their underlying metabolic pathways in tolerating water stress. USDA ARS anticipates finding novel metabolic biomarkers for breeding drought tolerant cultivars that will help support profitable lettuce production under water deficit conditions.

2.0 Scope

Analyze metabolites from 800 leaf samples in a lettuce population.

3.0 Tasks

USDA‐ARS at Salinas has a research requirement for the extraction and analysis of metabolites for 800 lettuce leaf samples. The non‐targeted metabolomics analysis includes profiling and relative quantification of about 1,500 metabolites by UPLC‐MS/MS. We will receive the data files, which inform us of the metabolites, the pathway of each metabolite, and the relative quantification in the sample.

4.0 Delivery/Period of Performance

The results of the metabolome analyses (metabolites, pathways, and quantification) shall be delivered to:

The Period of Performance is no more than 16 weeks from time of award. We are growing 800 lettuce plants during the fall and winter seasons of 2022. USDA‐ARS will send all samples to the contractor in February 2023.

7.0 Place of Performance

The extraction and analyses of metabolites will be performed at the vendor’s facility.
