Attachment A - Statement of Work General:

Contractor shall provide all services, supplies, equipment, and facilities necessary to provide the line items specified in accordance with the terms, conditions, and specifications contained in this document.

Background / Work to be performed.

Genomic DNA will be prepared in adequate quantity and quality for library preparation by ARS and shipped to the vendor. Vendor will construct libraries for both 16s (bacteria) and ITS (fungal) amplicons including blocking of contaminant (chloroplast, mitochondria, host) sequences with peptide nucleic acids, confirm library quality/quantity, normalize library pools and confirm normalization using an Illumina MinISeq platform using 2x150 paired-end sequencing chemistry, and sequence library pools on an Illumina MiSeq using V2 2x250 paired-end sequencing chemistry. Sequence data will be de-multiplexed, merged, trimmed, quality-filtered, resolved to taxonomic classifications using the DADA2 pipeline, and delivered to ARS by providing a link from which the sequence data and resulting analysis can be downloaded.

Review of Deliverables Table below is for supply- Ensure that there is a delivery location and any special instructions. Also, a POC that will receive the products. If there are any additional warranties, please note.

Quantity Product # Description Unit of Purchase Cost

Paired DNA sequence files for both 16s and ITS, taxonomic assignment files, metadata files.

DELIVERABLE SCHEDULE: Sequencing data should be returned within six to eight weeks of submission.

Method of Delivery: Sequence data and will be de-multiplexed and delivered to ARS by providing a link from which the sequence data (in .fastq format) along with output files from the bioinformatic analysis can be downloaded

Below is for Service.

Bacterial and fungal amplicon library preparation from 863 genomic DNA samples with standard quality and quantity confirmation and including blocking of contaminant (chloroplast, mitochondria, host) sequences with peptide nucleic acids, library normalization and pooling, confirmation of normalization on the Illumina MinISeq platform using 2x150 sequencing chemistry, paired-end sequencing on the Illumina MiSeq platform using V2 2x250 paired-end sequencing chemistry, complete bioinformatic analysis which includes both bacterial and fungal taxonomic classification of amplicons using the DADA2 pipeline.

Delivery Location:

USDA Agricultural Resource Service (ARS), Appalachian Fruit Research Station (AFRS) 2217 Wiltshire Road Kearneysville, WV 25430 erik.burchard@usda.gov

Period of performance- Date of award to 09/30/2021

INVOICES: To invoice, a vendor must first enroll in the Invoice Processing Platform (IPP) by visiting https://www.ipp.gov/vendors/index.htm. Once enrolled, all invoices must be submitted electronically through IPP. The IPP is a government-wide secure web-based payment information service offered free of charge to government agencies and their suppliers by the U.S. Department of Treasury’s Financial Management Service (FMS).

One-time enrollment in IPP means that you will receive a series of e-mails from Treasury services. The first email will have the IPP Logon ID and link to the IPP application. A second e-mail, containing the password will be sent within 24 hours. Once you receive these emails, please login to the IPP application and complete the registration process.

Benefits of registering with IPP include the ability for your company to create invoices directly from a contract award and submit them electronically, as well as:

• E-mail notification when invoice(s) are paid

• Online payment history

• Remittance download E-mail notifications of payments are sent when a payment is distributed to your bank account and will include all pertinent payment information. The IPP Customer Support Desk is available to assist users Monday through Friday (excluding bank holidays) from 8:00AM - 6:00PM ET, including answering any questions related to accessing IPP or completing the registration process.

Their toll-free number is (866) 973-3131 or they can be reached at: IPPCustomerSupport@fms.treas.go
