About this file

This is a Statement of Requirement for a USDA research project evaluating nasopharyngeal colonization of Mannheimia haemolytica bacteria in cattle. The project aims to assess how inactivation of specific bacterial regions (including capsular, sialic acid, filamentous hemagglutinin, leukotoxin, and adhesin features) affects colonization of wildtype and isogenic mutant strains, with particular focus on colonization during concurrent viral infection. The research addresses bovine respiratory disease, the most costly disease-related issue in the U.S. cattle industry, which is currently controlled primarily through antibiotic use rather than effective vaccination.

The contractor must procure and deliver 15 Holstein or Holstein-cross calves to USDA-NADC at 1920 Dayton Avenue, Ames, Iowa 50010, with delivery between mid-April and mid-May 2026. Calves must be 7-8 weeks old, sourced from farms within 100 miles of Ames, and meet extensive health requirements including BVD-negative status, Mycoplasma bovis-negative status, colostrum deprivation, deworming, freedom from ringworm, and demonstrated absence of clinical illness. Additional specifications include Certificate of Veterinary Inspection, sound legs and eyes, comparable sizing across the group, individual housing capability, and no milk replacer for the first 48 hours. The contractor may need to source more than 15 animals to ensure 15 meet all specifications, with pre-purchase bloodwork available to verify BRSV and Mycoplasma titer levels. All communication regarding animal health status and logistics must occur between the vendor and the Animal Resources Unit in Ames, with the government retaining authority to exclude non-compliant animals prior to arrival.

Statement of Requirement

Project: Mucosal colonization of Mannheimia haemolytica and isogenic mutants in cattle.

GENERAL INFORMATION

1.0 Scope of Work: Our objective is to evaluate the nasopharyngeal colonization of a wildtype Mannheimia haemolytica and seven isogenic mutants (capsular, sialic acid, filamentous hemagglutinin, leukotoxin, and three adhesin mutants) in cattle.

Determine what role, if any, inactivation of specific regions of Mannheimia has on the observed high-level colonization of wild type Mannheimia; the effect on colonization during a concurrent virus infection.

2.0 Background: Bacterial bovine respiratory disease continues to be the most costly disease-related issue facing the US cattle industry. Control of the disease is currently accomplished primarily through the extensive use of antibiotics. Vaccine control measures are widely available but suffer from relative inefficacy, in part due to problems with timely delivery. Bacterial capsule is prominent among these features, inhibiting phagocytic activity and complement-mediated killing. Sialic acid decoration of LPS appears to be an immunologic stealth mechanism for P.

multocida (and other organisms) whereby the bacterium can escape immune detection.

CONTRACTOR REQUIREMENTS

3.0 Technical Requirements/Tasks: This project requires the purchase of 15 Holstein or Holstein/cross calves. For this project we require the following of the calves:

1) Must be BVD-negative for antibody and antigen

2) Must be Mycoplasma bovis negative

3) Colostrum Deprived

4) No milk replacer for the first 48 hours

5) Housed individually

6) Must be dewormed

7) Must be free of ring-worm

8) Must be sound of foot and body

9) Must be 7-8 weeks old

10) Must be available for delivery Mid-April to Mid-May 2026

11) Animals must be transported directly to NADC with no other animals in the trailer.

12) Must have 15 calves available with comparable size across the group of the same breed (mixed dairy/beef breed)

13) Certificate of Veterinary Inspection required on all animals.

14) Calves shall demonstrate no signs of clinical illness.

15) Calves shall have sound legs and good eyes.

16) Calves must be sourced from a farm within 100 miles of Ames to limit transport time of the animals on the trailer.

17) Pre-Purchase blood work will be requested from the time of award until the arrival of animals if needed to check for BRSV and Mycoplasma titer level.

18) Communication with any possible vendor from the time of solicitation to the arrival of animals must be had between Animal Resources Unit in Ames and the possible vendor. This communication may include group or individual animal health status, pre-purchase blood work results, housing and feeding information that could be subject to change within this time frame.

19) Animal Resources Unit and the funding unit may exclude calves prior to arrival that do not meet the SOW requirements.

20) To obtain 15 animals that meet the SOW specifications; the vendor may need to obtain a group larger than 15 to allow for alternate animals to replace any that become sick or do not meet the health requirements.
