About this file
This document is a specifications sheet for a federal contract opportunity with the USDA for reagent kits compatible with the Rhapsody high-throughput microwell system. The procurement requires reagents and protocols capable of multiplexing porcine nuclei for single-nuclei whole-transcriptome and ATAC-seq analysis on the BD Rhapsody Scanner and HT-Express System, with capacity to run up to 8 lanes of samples simultaneously.
The contractor must provide reagent kits specifically compatible with BD Rhapsody microwell-based cartridge technology featuring multitier barcoding, explicitly excluding fluidics or droplet-based systems. The kits must support both whole transcriptome analysis (WTA) and ATAC-seq assays on the same cell or nuclei with multiplexing capability, and must accommodate a wide range of cell and nuclei quantities from hundreds to approximately 500,000 per cartridge. The required items include: BD Rhapsody Enhanced Cartridge Reagent Kit v3 (Qty 2), Rhapsody cDNA Kit (Qty 2), Rhapsody ATAC-Seq Tagmentation Kit (Qty 2), ATAC-seq Multiomics Amplification Kit (Qty 1), and Whole Transcriptome Analysis Kit (Qty 1). All items must be delivered to USDA, 1920 Dayton Avenue, Building 24, Ames, Iowa 50010 within 15 business days of award date, or on a delivery schedule mutually agreed upon by both parties.

Specifications
Project: Reagents for use with the Rhapsody high-throughput (HT) microwell system for multiplexing porcine nuclei for single-nuclei whole-transcriptome and ATAC-seq analysis.
CONTRACTOR REQUIREMENTS
Specifications
• Contractor must be able to provide all reagents and protocols compatibile with the Rhapsody Scanner and HT-Express System to run up to 8 lanes of samples at one time (and two samples per lane with use of cell tags compatible with swine nuclei)
• Reagent kits must be compatable with BD Rhapsody microwell-based catridge technology with a multititier barcoding system. The reagents must be for a system that is microwell/catridge based and NOT fluidics/droplet based.
• Contractor must be able to provide reagent kits compatible with both whole transcriptome analysis (WTA) and ATAC-seq assays on same cell/nuclei with the ability to multiplex nuclei samples.
• Contractor must provide reagent kits capable of processing a wide range of cells/nuclei on one catridge (from hundreds to approximately 500,000).
