Statement of Work Procure Tristar 3 Multimode luminescent, absorbance and fluorescent detection instrument
BACKGROUND:
The National Institute of Health (NIH) mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability. The National Human Genome Research Institute (NHGRI) vision is to improve the health of all humans through advances in genomics research. The Collins laboratory has developed and mouse model for Hutchinson-Gilford Progeria Syndrome (HGPS), that faithfully reproduces the cardiovascular phenotype and pathology as well as the premature death characteristics of the HGPS subjects. Key features in common are vascular smooth muscle depletion in the large arteries, vascular extracellular matrix deposition, calcification of the aortic vessel wall, cardiac muscle fibrosis as well as cardiac strain. Although mortality is due to myocardial infarction or stroke due to rapidly progressive atherosclerosis, cells and tissues derived from common mesenchymal progenitors are particularly susceptible to the pathology that results from progerin accumulation. While progressive bone dysplasia is one of the defining phenotypic features of HGPS, several studies suggest epidemiologic and biologic links between aging-related cardiovascular disease and osteoporosis.
Purpose and Objectives:
The purpose of this acquisition is to procure Tristar 3 Multimode luminescent, absorbance and fluorescent detection instrument for NHGRI--Collins Laboratory for clinical study (HGPS ZIA HG200305). To support this research the scientific investigators aim to employ the Berthold Tristar 3 Multimode Reader capable of luminescent, absorbance and fluorescent detection of specific indicators of gene expression and calcium flux in cell lines. This specific instrument will allow researchers to directly extrapolate previously acquired data from a now-defunct instrument to future experiments.
Procurement:
To procure the following:
1. Item Number: 69173-20: Tristar 3 for UV/VIS Abs/Lum/FL/Temp control/Cooled PMT/ICE-software, LB 942 Tristar 3 Multimode Microplate Reader with reading technologies: absorbance, luminescence, fluorescence intensity optimum performance and sensitivity due to unique optical system "ONE-4-ALL" excitation/emission filter slider with 5 positions, Quantity: One Each;
2. Item Number: 54116-32: injector #2 measurement position, 10-100µl, Quantity: One Each;
3. Item Number: 54116-33: injector #3 measurement position, 10-100µl, Quantity: One Each;
4. Item Number: 38470: cwl 430nm, fwhm 10nm, 60%, 25mm Excitation CFP, Quantity: One Each;
5. Item Number: 39798: cwl 495nm, fwhm 10nm, 70%, 25mm Emission CFP, Quantity: One Each;
6. Item Number: 55140: cwl 350nm, fwhm 50nm, 50%, 25.4mm Excitation FURA-2 Calcium Free, Quantity: One Each;
7. Item Number: 40094: cwl 510nm, fwhm 40nm, 75%, 25mm Emission FURA-2 Calcium Free, Quantity: One Each; and
8. Item Number: IN-1: Installation and Training.
9. Shipping shall be paid upon delivery.

Contractor’s Requirement:
· The Contractor shall deliver the Tristar 3 Multimode luminescent, absorbance and fluorescent detection instrument.
· The Contractor shall provide sensitivity:
· i. Luminescence: <6 amol ATP; <3 zmole firefly luciferase;
· Fluorescence: < 0.3 fmole FITC ; and
· Absorbance: 0 - 3.5 OD @ 400 - 700 nm (0 - 3 OD @ 340 nm) precision <0.6 % cv (@ 2.5 OD) accuracy <2 % cv (@ 2.5 OD) Spectral range 340-700nm.
· The Contractor shall provide low crosstalk using crosstalk reduction design <5 x 10-6.
· The Contractor shall provide up to 3 internal bellows type reagent “JET” injectors. Two are in the Luminescence measurement position, one in the pre-measurement position (luminescence) and one in the fluorescence measurement position The Injectors are freely adjustable from 10ul to 100ul volume and can be used with 384 well applications. Not to be syringe type which tend to clog and wear out faster than a bellows type injector.
· The Contractor shall provide system with “JET” injection technology, the injectors combine high speed and reliable mixing of reagents with outstanding precision into 96 and 384 well plates. >99% precision, >99% accuracy with a CV of <3%.
· The Contractor shall provide ICE software.
· The Contractor shall provide: 5-position emission filter slide and 5-position excitation filter slide; Optional Temperature control with cooled PMT, plate heating +5°C above RT and 45°C; Standard plate height adjustment to accommodate 6-384 well plates.
· The Contractor shall provide: software control of priming and cleaning of injectors; Low dead volume priming of ~800uL; unique “UNLOAD” feature returns >60% of the reagent to the reagent vial, this helps save expensive reagents.
· The Contractor shall provide: Agitation Function: Orbital, Double Orbital, and Linear; small footprint; dimensions 330mm W X 270mm H X 430mm D.
Government Property:
No government furnished property will be supplied to the contractor.
Government Responsibilities.
· The Government shall receive and inspect equipment.
· The Government shall review and approve all invoices.
· The Government shall at all times keep the Equipment in the environmental conditions recommended by the manufacturer. The instrument and all associated hardware will be protected from adverse elements, such as dirt, dust, and liquids of any kind.

Acceptance criteria:
Upon receipt of the goods shipped hereunder the Government shall inspect them immediately and shall within five (5) days give the Contractor written notice of any claim that the goods do not conform with the terms of the order. Should the Government fail to give such notice, it shall be deemed to have accepted the goods and shall be bound to pay for them. The Contractor expressly waives any rights it may have to revoke acceptance after such five (5) days.
If Applicable: NIH Facility Access Requirements due to COVID-19:
The Contractor should follow required safety protocols consistent with their vaccination status while accessing the facility. For a NIH owned facilities and some NIH leased facilities, maintenance, and other short duration contractor personnel (non-transient services) will be treated as visitors. Visitors will attest to vaccination or show proof of negative testing within the previous 72 hours when they are processed through NIH visitor entrances and should follow required workplace safety protocols consistent with their vaccination status while accessing a Federal facility. All Contractors and their contractor personnel that will be processed as visitors to NIH facilities should be familiar with the NIH Visitor Guidance and must follow the safety requirements as outlined in the NIH Safety Guidance in accordance with the Centers for Disease Control (CDC) COVID-19 Community Levels (link is external) in effect at the time of facility access. NIH Visitor and safety guidance are subject to change and should be reviewed frequently for updates.
