About this file
This is a Statement of Work (SOW) for advanced genetic sequencing services supporting a VA research study on Alzheimer's Disease. The Department of Veterans Affairs seeks a contractor to perform next-generation sequencing on mouse brain and lung samples, focusing on analyzing molecular changes from high-fat diet and environmental toxin exposures. The project involves processing, sequencing, and quantifying RNA from samples using single-cell RNA sequencing (scRNA-seq) on the PARSE platform, with an anticipated 4-20 samples per year.
The contract will be a firm-fixed price for a 5-year period from June 30, 2025, to June 29, 2030, with a solicitation number of 36C24E25Q0045. The contractor must provide comprehensive genetic sequencing services, including RNA library preparation, sequencing, and a data report detailing identified and quantified RNA transcripts within 30 days of sample receipt. Quotes are due by 11:59 EST on June 11, 2025, and will be evaluated on technical capabilities, past performance, and price. The contract will be awarded through full and open competition with the NAICS code 541715 for Research and Development in Physical, Engineering, and Life Sciences.

STATEMENT OF WORK
A. GENERAL INFORMATION
1. INTRODUCTION/BACKGROUND:
Our objective is to determine key features that differentiate the spatial configuration and expression profile of mouse brain from either WT or Alzheimer’s Disease expressing (AD) mice with confounding factors such as high-fat diet or environmental toxin exposure. We may identify new targets for AD treatment by identifying two new mechanisms of disease progression: 1) Enhanced neuroinflammation due to exacerbation of inflammatory factors due to high-fat diet or environmental toxin exposure in the AD background. 2) Identification of new disease mechanisms may improve the care of Veterans with AD, high-fat diet, and/or environmental toxin exposure thereby decreasing costs for the Veterans Affairs Health Care System.
2. OBJECTIVE:
We seek a contractor to perform next-generation sequencing on our various mouse brain and lung samples for our study Impact of Obesity on Brain Immune Response and Cognition 1594806. These samples will consist of either spatially segregated mouse brain and lung or single-cell suspensions. Samples will consist of male and female mice either sham exposed (control) or given high-fat diet (experimental) or burn pit material (experimental) or both. These experiments will reveal the molecular changes caused from these exposures.
3. MAJOR REQUIREMENTS:
Contractor shall be able to process, sequence, and quantify the RNA from our samples on a massive scale (in reference, specifically, to the depth and complexity of data generated by single-cell RNA sequencing (scRNA-seq, not the overall number of samples) with high-accuracy, dependability, and reproducibility. Although the VA anticipates sending 4 to 20 samples per year, each sample contains thousands of individual cells. Each of these cells is individually profiled, generating extensive data that can exceed 100 million sequencing reads per sample. Therefore, while the sample count is modest, the scale of the transcriptomic data and analytic demand is significant.
The specific task the contractor shall perform involves receiving samples, performing RNA library preparation, sequencing of the RNA, and preparing a data output including sequence identity and quantity for all samples.
Contractor shall provide start-to-finish advanced genetic sequencing and quantitation services tailored to the analysis of single-cells. “Start-to-finish” includes the comprehensive workflow required for single-cell RNA sequencing using the PARSE platform, to include RNA tagging, library construction, sequencing, and bioinformatics. This workflow is distinct from bulk RNA-seq and requires specialized expertise and platform compatibility. Contractor laboratory must be able to provide accurate and reproducible quantitative results for a variety of tissue types and sequencing modalities. Data generated in this project will be owned by the VA. After the contract is completed, the contractor shall return any unused samples to the VA. The Government will provide the funding for the transfer and services provided by the Contractor. The lab anticipates sending 4 to 20 samples per year depending on experimental outcomes.

8. REPORTS/DELIVERABLES:
Contractor will receive mouse lung and brain samples and deliver a data report detailing the RNA transcripts that were identified and quantified. Data report shall be provided within 30 days after Contractor confirms receipt of the samples.
