About this file

This document is a Purchase Description for an Automated Cryo-EM Sample Preparation Instrument, issued by the Structural Biology Core (SBC) of the Vaccine Research Center at the National Institute of Allergy and Infectious Diseases (NIAID), National Institutes of Health (NIH). The procurement includes one Chameleon Cryo-EM Sample Preparation Instrument with specific accessories, such as two Cryogen Bowl Assemblies, an Ethane Condensing Kit, a Desktop PC Controller, and a Consumables Starter Pack. The instrument is designed for high-resolution cryo-electron microscopy sample preparation, with capabilities including sample temperature control (4°C to 40°C), batch capacity of 8 grids, and integrated features like liquid nitrogen level monitoring and a closed-loop glow discharge module.

The purchase description specifies delivery within 30 days after receipt of order and includes a 12-month warranty covering replacement of defective components due to manufacturing or design defects. The instrument will support the SBC's research in developing antibody therapeutics and vaccines for infectious diseases, including COVID-19, HIV-1, malaria, flu, and RSV, by enabling more consistent and higher-quality sample preparation compared to manual plunging techniques. The total quantity requested is one instrument with associated accessories and consumables, tailored to meet the specialized needs of the Vaccine Research Center's structural biology research.

PURCHASE DESCRIPTION

1. Statement of Need and Purpose: Automated Cryo-EM Sample Preparation Instrument

2. Background Information and Objective: The Structural Biology Core (SBC), Virology Laboratory (VL), Vaccine Research Center (VRC), National Institute of Allergy and Infectious Diseases (NIAID), National Institutes of Health (NIH) uses a structure-based approach for developing antibody therapeutics and vaccines for infectious diseases, including but not limited to COVID-19, HIV-1, malaria, flu, and RSV. The structural analysis of pathogen proteins, antibodies, and vaccines is essential to current and future SBC research. It requires high-resolution cryo-EM data attainable only with high-quality samples. Currently, SBC relies on manual plungers for the preparation of cryo-EM grids, which can yield suboptimal results, particularly with challenging samples. The adoption of automated, non-blotting technology offers a promising solution to this issue. To meet the specialized needs of SBC VRC, SBC needs to acquire the Chameleon Cryo- EM Sample Preparation Instrument.
