About this file
This document is a Specifications file for a procurement of scientific laboratory equipment by the Agricultural Research Service (ARS).
The ARS Sustainable Perennial Crops Laboratory in Beltsville, Maryland seeks to purchase a nano-fluidic/microfluidic genotyping and real-time Polymerase Chain Reaction (qPCR) system to support a 5-year research project focused on genotypic characterization of genetic resources for cacao, coffee, and other tropical perennial crops. The research objective is to identify the genetic basis of genetic diversity in these crops to support breeding programs that develop high-quality, disease-tolerant varieties. The system must be a complete, ready-to-use benchtop instrument capable of high-throughput analysis and must include a dedicated computer with analysis software, calibration reagents, and on-site training. Technical specifications require the system to accommodate multiple plate sizes (48X48, 96X96, 24X192, and 192X24 well plates) with nanofluidic volume reactions, operate at nano-liter scale reaction volumes, and feature Peltier-based thermal cycling from 4 to 99°C with ramp rates up to 5.5°C per second. The optical system must include fluorescence excitation capabilities at 485, 530, and 580 nm with emission detection at 525, 570, and 630 nm, a 20-megapixel CMOS camera, and compatibility with a wide range of dyes and chemistries including the ability to detect up to three fluorescent dyes per assay with support for both probe-based and DNA-binding (intercalating) dye chemistry. The instrument dimensions are specified at 64.2 x 26.0 x 53.5 cm with a weight of 37 kg.

SPECIFICATIONS
I. BACKGROUND
The ARS Sustainable Perennial Crops Laboratory (SPCL) in Beltsville, Maryland, plans to purchase a nano-fluidic/microfluidic genotyping and real-time Polymerase Chain Reaction (qPCR) system to support the 5-year research project, " Genotypic Characterization of Genetic Resources for Cacao, Coffee, and Other Tropical Perennial Crops Economically Important to the United States " (Project No. 8042-21000-303-00-D). The core objective of this research is to identify the genetic basis of genetic diversity in cacao and coffee to support the improvement of breeding programs to develop high quality, durable disease-tolerant varieties.
To achieve these goals, the research requires precise and reliable quantification of genetic diversity through genotyping. The BioMark X9 system is fundamental for identifying and validating single nucleotide polymorphism data, screening resistant plant candidates, and analyzing plant genetic populations. To handle the large volume of samples and the large quantity of data generated a highly efficient nanofluidic instrument must be capable of high-throughput analysis.
The goal of this research is to generate high-quality, reproducible data that will serve as the foundation for developing new superior tropical tree crops to supply safe and stable supplies of these commodities to the USA market. To accelerate research progress, the instrument must be a complete, ready-to-use system that can be linked to the existing data flow and includes a dedicated computer with analysis software, all necessary calibration reagents, and on-site training.
II. REQUIREMENTS
The real-time PCR system must be capable of the following:
• System Type: Benchtop nanofludic genotyping and real-time PCR instrument.
• Size & Weight: 64.2 x 26.0 x 53.5 cm (approximately 25.2 x 10.2 x 21.1 inches), 37 kg (approximately 83 lbs)
• Sample Format & Throughput:
Must accommodate multiple plate size 48X48; 96X96; 24X192;
192X24 well plates with nanofluidic volume reactions.
Reaction Volume: Nano Liter scale volumes.
• Thermal Performance: Peltier-based thermal cycling (4 to 99ºC) with excellent thermal uniformity and accuracy across the block to ensure reproducibility (ramp rate up to 5.5º C/sec).
• Optical System & Performance:
Excitation: Fluorescence excitation 485, 530 and 580 nm; Fluorescence emission 525, 570 630 nm Detection: CMOS camera 20Mpixels.
