Statement Work (SOW)

1. Background

The Food and Drug Administration (FDA) is responsible for protecting the public health by ensuring the safety, efficacy, and security of human and veterinary drugs, biological products, and medical devices; and by ensuring the safety of our nation's food supply, cosmetics, and products that emit radiation. The FDA also has responsibility for regulating the manufacturing, marketing, and distribution of tobacco products to protect the public health and to reduce tobacco use by minors. Additionally, the FDA is responsible for advancing the public health by helping to speed innovations that make medical products more effective, safer, and more affordable and by helping the public get the accurate, science-based information they need to use medical products and foods to maintain and improve their health.

The organization sponsoring and supporting this requirement is the Office of Computational Science (OCS) in the (super) Office of Translational Sciences (OTS) under the Center for Drug Evaluation and Research

(CDER).

The Center for Drug Evaluation and Research (CDER) performs an essential public health task by making sure that safe and effective drugs are available to improve the health of people in the United States. As part of the U.S. Food and Drug Administration (FDA), CDER regulates over the counter and prescription drugs, including biological therapeutics and generic drugs.

The Office of Translational Sciences empowers a collaborative and high performing workforce to champion innovation and advance global human drug development. The vision of this super office is to drive advancements in human health through scientific and regulatory innovation. A summary of current work being done includes:

• Promoting scientific collaboration and innovation in drug regulatory review across CDER

• Assuring the validity of clinical trial design and analysis in regulatory decision making

• Developing and applying quantitative and statistical approaches to decision making in regulatory review process

• Ensuring alignment with CDER goals

• Serving the CDER scientific community in establishing technology transfer agreements that are vital to collaboration with the broader scientific community

• Maintaining knowledge management databases that can be the basis of improvements in the regulatory review process

• Overseeing bioequivalence inspections to ensure the availability of safe and effective generic equivalents of investigational drugs

The mission of the Office of Scientific Computing is to provide CDER reviewers innovative and reliable solutions that improve and strengthen the scientific review process by integrating data, tools, and training.

The focus of OCS’s current efforts include:

• Building solutions that drive New Drug Modernization, promote access to quality data, and optimize data analysis capabilities

• Processing Study Data for rapid use

• Delivering effective tools, technologies, services, and training to meet CDER’s mission

• Delivering solutions that streamline Bioresearch Monitoring (BIMO) workflow for reviewers and inspectors

• Spearheading the CDER Innovation Board to harness subject matter expertise across FDA

• Offering cloud-enabled innovation space for tools, technologies, and data exploration

• Connecting CDER staff to the scientific computing resources and technology needed to drive efficient analysis and cost-effective solutions mailto:info@JohnSnowLabs.com

• Collaborating with industry, academia, and other external partners through our work with PHUSE, CDISC and BioCelerate

Currently, the CDER process to identify submissions containing standard data is labor intensive: the Office of Business Informatics reviews all new submissions and performs an assessment on a few relevant files to determine if the submission appears to be SDTM (Study Data Tabulation Model) compliant. The DataFit system automatically and systematically checks submission and validates conformance of study data to CDISC Standards (e.g. SDTM, ADaM [Analysis Data Model], SEND [Standard for Exchange of Nonclinical Data]). This automated systematic approach will allow CDER to successfully transition to a more advanced regulatory process which takes full advantage of the benefits of high-quality standardized data improving the ability for Food and Drug Administration (FDA) reviewers to make science based regulatory decisions faster.

2. Project Scope

The objective is to install, update, support, and develop enhancements and to maintain DataFit, the CDER’s data standards validation platform. The Government will not reimburse the Contractor for any work not authorized by the CO, including work outside the scope of the contract.

All work shall be accomplished in accordance with the FDA, Department of Health and Human Services

(HHS), and Office of Management and Budget (OMB) standards described below. In the event the Contractor is unable to meet the standards, they shall immediately notify the COR. The Contractor shall comply with applicable federal information technology and information management laws, regulations, policies, and standards at the government‐wide, HHS, and FDA levels, pursuant to guidance and instruction from the COR and as provided in this statement of work. At the government‐wide level, these include OMB, National Institute of Standards and Technology (NIST), and General Accounting Office (GAO). These can be primarily found at or through the Federal CIO Council website at: https://www.cio.gov. See the underlying statement of work for all technical and conformance guidelines. The below standards and guidelines have been included for quick reference.

6.1 Software Development Lifecycle Standards

The Contractor shall follow the standard development lifecycle for FDA IT systems and modules. Currently, the FDA follows the HHS Enterprise Performance Life Cycle (EPLC) management process, which includes both waterfall and agile methodologies. FDA expects the use of agile methodologies. This process‐driven IT lifecycle management approach emphasizes enterprise integration based on development of sound business and technical requirements. The framework will evolve over time in response to changes in technology and business needs and, therefore, the Contractor is expected to adapt accordingly to comply with the most current framework methodology in performance of work under this statement of work. The Contractor shall consult with the COR and/or COR designee to clarify the EPLC artifacts required for an individual project or system. The required artifacts depend on the release type and are tailored through the Project Process Agreement (PPA). Furthermore, the Contractor shall ensure that updated artifacts are maintained in the appropriate repositories.

Additional requirements for IT application design, development, or support include the following:

A. The Contractor (and/or any subcontractor) shall ensure IT applications designed and developed for end users (including mobile applications and software licenses) run in the standard user context without requiring elevated administrative privileges.

B. The Contractor (and/or any subcontractor) shall follow secure coding best practice requirements, as directed by United States Computer Emergency Readiness Team (US-CERT) specified standards and the Open Web Application Security Project (OWASP), that will limit system https://www.cio.gov./ software vulnerability exploits.

C. The Contractor (and/or any subcontractor) shall ensure that computer software developed on behalf of HHS/FDA or tailored from an open-source product, is fully functional and operates correctly on systems configured in accordance with government policy and federal configuration standards. The contractor shall test applicable products and versions with all relevant and current updates and patches updated prior to installing in the HHS/FDA environment. No sensitive data shall be used during software testing.

D. The Contractor (and/or any subcontractor) shall protect information that is deemed sensitive from unauthorized disclosure to persons, organizations, or subcontractors who do not have a need to know the information. Information which, either alone or when compared with other reasonably-available information, is deemed sensitive or proprietary by HHS/FDA shall be protected as instructed in accordance with the magnitude of the loss or harm that could result from inadvertent or deliberate disclosure, alteration, or destruction of the data. This language also applies to all subcontractors that are performing under this contract.

6.2 Release Management

The Contractor shall work within the FDA guidelines for Release Management and Coordination. This process provides application and service teams a shared solution for planning releases. The Release Management and Coordination meeting will complement the Change Control and Configuration Management process by:

• Forecasting and communicating upcoming releases,

• Assisting with the resource confirmation process, and

• Expediting the Change Control Board meetings.

The Release Management activities occur before requests for change are submitted to the Change Control Board.

6.3 Change Control and Configuration Management

The FDA has a Change Control, Configuration and Release Management Team who is responsible for managing the process of formally changing the FDA IT Infrastructure by facilitating the Change Control Board and Configuration Items database. Documentation and procedures will be provided by COR at award.

The team performs the following key functions:

• Facilitate the Change Control Board (CCB).

• Is Process Champion for Configuration and Change Management, including EPLC artifacts.

• Administers the automated Configuration Management Tools.

The FDA’s Configuration Management process is used to control changes within the functional and physical aspects of FDA systems, thus ensuring the proper maintenance of integrity and traceability throughout the system development life cycle.

6.4 Request Quality Services and Technology from IT (RQST-IT Tracker) The FDA has instituted the RQST‐IT process to track projects, provisioning, technologies and configuration changes across the agency. The introduction of a new technology or IT project, version change of an already approved technology or change to the configuration of the system requires the submission and approval of an RQST‐IT request. The Contractor shall be responsible for supporting the RQST‐IT process including writing and submitting requests and tracking them from inception through completion.

The FDA Center IT Investment Review Boards (ITIRBs) may need to be included in a coordinated review process. The RQST‐IT process is also used to request the provisioning of new servers in both data centers.

6.5 Data Quality Standards

FDA has approved data standards for e‐submissions and their regulatory review process. For example, CDER data standards references and resources can be found at the following site:

https://www.fda.gov/forindustry/datastandards/studydatastandards

The CDER Data Standards Program does the following:

• Identifies data standard needs.

• Determines priorities for these needs.

• Works with organizations that develop standards, industry, and other stakeholders to find or develop the necessary standards.

Where current standard specifications are unavailable or inadequate, CDER plans to engage appropriate Standards Development Organizations (SDOs), as well as industry and other stakeholders, to develop new standards through SDO processes.
