Statement of Needs
The Leica Stellaris 5 system will be equipped with a White Line Laser (WLL), will provide the following:
• Eight (8) simultaneous excitation lines between 485nm and 685nm
• Each line tunable in 1nm increments
• Dynamic beam splitter AOBS® instantly adapts to each laser line (up to 8 simultaneously)
• Pulsed laser for lifetime imaging with all Power HyD® detectors
• Three (3) Simultaneous Spectral SP® Detection Channels based on Leica's High-Effifficiency Pellin-Broca Prism (upgradeable to 5 channels)
• Power HyD® S, Positions 1, 2, and 3
◦ New silicon-based, solid state detector with >56% Photon Detection Efficiency (PDE)
◦ Increased sensitivity in the NIR up to 850nm
• >2-fold increase in dynamic range over previous generation HyD detectors
◦ Analog and Photon Counting modes with PowerCounting
• TauSense Family of modalities that incorporate a novel use of fluorescence lifetime information
• TauContrast - generate high-contrast, detailed images using lifetime information for functional imaging, including cell signaling, metabolic state, pH or ion concentration
• TauSeparation - Multiplex beyond spectral options. Separate spectrally overlapping fluorophores to acquire 2 or more channels using 1 laser line, 1 detector, and 1 scan
• TauGating - Unveil information masked by auto-fluorescence and eliminate reflection at the coverglass
• Premium CUDA Workstation with high-end GPU for accelerated 3D image display and processing
• Leica Lightning for Near Real-Time Confocal SuperResolution and Image Detail Extraction (S/N improvement)
• SuperResolution with resolution to 120nm lateral and 200nm axial
• Lighting & Dynamic Signal Enhancement with Conventional and Resonant Scanning for maximum S/N and SuperResolution without compromising Speed or FOV
• Compatible with all microscope objectives
