Version 11-30-2016-1 FAR Part 11—Describing Agency Needs
PURCHASE DESCRIPTION
1. Statement of Need and Purpose:
The study of vascular function is central to understand the neurovascular biology. The Neurovascular Research Unit at the National Institute of Neurological Disorders and Stroke (NINDS) needs non-invasive or minimally invasive techniques to investigate the vascular structure and function of cerebral vasculature. Important and powerful ex-vivo methodologies, i.e. arteriography, have been developed to help better understand the complex, yet vital, nature of blood vessel biology. The NRU will use arteriography (pressure and flow) to study physiological vascular response in different cerebral and peripheral blood vessels. Pressure arteriography is the gold-standard for quantifying function, reactivity, and mechanics in isolated perfused blood vessels. Pressure arteriography is a powerful tool and comprises a number of different hardware and software components.
2. Background Information and Objective:
One of the aims of the Neurovascular Research Unit is to perform ex-vivo vascular functions measurement. In order to conduct these experiments, the NRU will utilize arteriography with pressure and flow manipulations which will allow it to monitor and quantify vasoactive responses in small blood vessels. This technique allows the study of isolated vascular segments from different animal models with physiological manipulations and pathophysiology models of Vascular Cognitive Impairment and Dementia (VCID), including Alzheimer’s Disease and Stroke.
3. Generic Name of Product: Pressure Arteriography and Pressure + Flow Arteriography
4. Purchase Description: Living Systems Instrumentation (LSI) Pressure Arteriography and Pressure + Flow Arteriography (Brand Name Only)
5. Salient characteristics
All the items below shall be part of a modularized system to allow for easy cleaning and maintenance by the end-user. The components below shall be able to be switched out in the event of a failure to allow the system to come back into operation quickly.
Pressure Servo Controller with Peristaltic Pump
The pressure servo controller with peristaltic pump is used to precisely control the intravascular pressure in isolated cannulated blood vessels. Many other uses are also possible, including maintaining pressure in isolated organ beds, gastrointestinal tissue, urinary tract, or lymphatics. This instrument is used for examination of myogenic responsiveness of isolated cannulated blood vessels.
System shall achieve either a constant pressure (pressure mode; 0 to 200 mmHg) or a constant flow rate (flow mode; 3 µl/min to 2.4 ml/min, depending on tube set used).
In the pressure mode of operation, a flow-through pressure transducer placed in-line with the blood vessel, or other biological preparation, shall sense the pressure and maintain pressure at the user-specified set-point. In the flow mode of operation, the peristaltic pump shall supply a user-defined constant flow rate, and the pressure transducer monitors perfusion pressure. Pressure is indicated directly in mmHg on a digital panel meter in both modes, and is available as an analog output voltage for data acquisition. Pressure or flow set-points need the ability to be programmed through an external voltage signal applied to the instrument.
Thermal Circulating Bath
The thermal circulating bath is a heating circulator that shall include the immersion heater unit, water bath, and circulating pump. The unit shall have a small footprint and low profile.
Two Channel Superfusion Peristaltic Pump
The peristaltic pump is for vessel bath superfusion. The pump shall include two flow heads, one for delivering solution to the bath (inflow), and one for removing fluid from the bath (outflow). The pumps shall be isolated from the vessel chamber, so they don’t impart any vibration on the vessel being studied.
Vessel Dimensioning Acquisition
Vessel dimensioning acquisition is a module for those performing experiments with isolated perfused vessels. It provides reliable calculation and reporting of vessel measurements, offering valuable features such as multiple ROIs, line averaging, and auto-gained contrast. Item shall be able to be paired with any DAQ system (iWorx, LabChart, etc.) to record the data.
Pressure Perfusion Monitor
The pressure perfusion monitor is used for isolated, perfused vessel studies to aid the investigator in ensuring that a perfused vessel is exposed to a desired physiological pressure. The device shall have two pressure input channels, enabling the measurement of upstream and downstream pressure, average pressure, and pressure differential across a cannulated blood vessel. It can also be used for measuring the pressures at two locations in a vessel bed or isolated tissue. In conjunction with the pressure servo controller, a transmural vessel pressure or a perfusion pressure may automatically be maintained during perfusion.
The item shall feature four analog voltage outputs, which can be used to record pressure on both pressure channels, the pressure differential between the two pressure inputs, and the average of both pressure inputs.
Flow Control Peristaltic Pump
The miniature peristaltic pump shall deliver precise volume flow rates over a range determined by the setting of two calibrated potentiometer controls, and the tube set size chosen.
Flow Indicator
The flow indicator shall provide a digital readout of the flow rate delivered by the FC flow control pump or the pressure servo control pump. Flow rate shall be monitored directly on the front-panel meter in µl/min, or using the analog voltage output signal, which is compatible with most data acquisition systems. The flow indicator shall also have a connector that enables remote control of flow rate using a D/A output or other analog voltage source.
Vessel Flow & Dimension Acquisition
Vessel flow and dimension acquisition is a module for those performing experiments with isolated perfused vessels. It shall provide reliable calculation and reporting of vessel measurements, offering valuable features such as multiple ROIs, line averaging, and auto-gained contrast.
The module shall use flow rate and inlet/outlet pressures to calculate vessel flow characteristics (vessel wall shear stress, vascular resistance, Reynolds Number) in real time.
Miscellaneous
All appropriate tubing, fittings, tube sets, pump stand, calibration kits, etc. shall be identified and quoted for the above items.
