PURCHASE DESCRIPTION
BRAND NAME OR EQUAL
Multiphoton Microscope
1. Statement of Need and Purpose:
The Spatial Navigation and Memory Unit needs a multiphoton microscope to perform research studies focusing on the synaptic plasticity in the medial entorhinal cortex (MEC) during spatial learning. Specifically, the microscope will be combined with two fixed-wavelength (920 nm and 1064 nm) compact femtosecond lasers to image synaptic surface dynamics of GluA1 using GluA1-SEP and calcium dynamics of the MEC neuron using jRGECO, respectively.
2. Background Information and Objective:
Spatial Navigation and Memory Unit (SNMU), which aims to investigate the neural basis pf spatial navigation and memory. Specifically, the experiments in the laboratory plans to use in vivo two-photon imaging approach to measure calcium dynamics of the MEC neurons while observing their structural and molecular properties at the subcellular level. The laboratory will require one multiphoton microscope to achieve the above measurement. The collected data are important to understand the synaptic plasticity of the MEC in spatial learning.
3. Generic Name of Product:
Multiphoton microscope
4. Purchase Description:
Vendor shall provide a system brand name or equal to the Bruker Nano Ultima Investigator DL Laser Scanning Microscope INV-02.
5. Minimum Salient characteristics The microscope system shall provide the following specifications:
• The microscope can perform off-axis imaging (15o to 35o in both clockwise and counterclockwise directions) without affecting the efficiency of two-photon excitation and signal collection. To ensure imaging stability, the off-axis imaging must be achieved by only rotating the objective mounted on a nosepiece without rotating the entire microscope body.
• The microscope is precisely controlled by a vendor-supported software Prairie View
• The microscope or similar microscopes from the same vendor should be used in at least five publications
• Performing both galvo and resonant galvo scanning
Additional features of the product:
• The microscope can be used with awake, head-fixed mice running on an air-floating ball. While ensuring the stability of the microscope, the distance between the bottom of the objective and the optical table should be about 350 mm.
• The angle of collection from back aperture of objective is 14 degrees.
• The microscope must have an option to upgrade collection path with two additional PMT channels for four-color imaging.
• The microscope contains 8kHz resonant galvanometer for high-speed scanning at 30 frames per second (512 × 512 pixels) with user-definable resolution and region of interest. Resonant scanner must be relayed to standard XY imaging galvos to provide flexibility to image in galvo mode or resonant mode and to re-center the resonant scan for dynamic ROIs in resonant mode.
• The microscope contains at least two GaAsP detectors combined with appropriate filter sets (changeable by users) for high sensitivity simultaneous detection of green and red fluorescence signals. GaAsP detectors must have built-in high-speed shutters.
• The microscope must allow the excitation of the sample at different wavelengths, such as 920nm and 1064nm.
• To maintain the center position of the mouse in virtual reality during off-axis imaging, the microscope body should be able to laterally translate for at least 30 mm.
• In order to focus the sample to the tilted objective, the software must control the movement of the microscope X, Y, Z motors together to move the objective along its optical axis and perpendicular to the sample surface.
• The microscope must be placed on an optical table (5 ft × 5 ft) and leave enough space for the two lasers and a virtual reality setup.
6. Quantity:
One system
7. Delivery, Installation, and Training:
The Contractor shall deliver and install the equipment within 90 days after receipt of order. Delivery must be Freight on Board (FOB) Destination and include inside delivery. On-site training must take place within two (2) weeks of equipment installation. The equipment shall be delivered and installed in coordination with the Contracting Officer’s Representative (COR). The contractor shall provide an Original Equipment Manufacturer (OEM) certified service technician to perform setup, installation, and an on-site training session for staff members. The contractor shall also provide one day of follow up training with Applications Specialist.
8. Warranty:
Contractor shall warrant that the Equipment will be free from material defects for a period of at least twelve (12) months from the date of acceptance. All service performed on this equipment shall be provided by certified and factory trained technicians. Technicians shall have the ability to diagnose the systems on site via remote access. Contractor shall provide any required replacement components due to malfunctions on-site. The Contractor shall demonstrate that it is capable of providing this level and speed of service. Warranty shall not include routine or periodic maintenance services.
The contractor shall provide software support for life of the instrument and one year of warranty covering parts and labor.
