About this file
This is a Purchase Description document under FAR Part 11 that outlines the National Institute on Aging's (NIA) procurement requirements for multiplexed precision proteomics immunoassay panels and bulk reagents for neurological disease biomarker research.
The NIA requires seven (7) NULISAseq Neuro 220 Panels and two (2) NULISA 10X Wash Buffer units to support an in-depth study of protein changes associated with neurodegeneration and the effects of dietary interventions on chronic pain patients. The research leverages samples from a randomized clinical trial examining fatty-acid dietary interventions and aims to identify plasma and exosome-derived biomarkers for monitoring treatment response and guiding precision medicine interventions in older adults with chronic pain. The proteomics panels must provide ultra-high sensitivity detection at attomolar levels (as low as 0.1 fg/mL), wide dynamic range up to 12 logs without dilution, and true high-multiplexing capability for 100 to 1,000s of targets in a single reaction. The panels must be compatible with the ARGO HT System, a fully automated all-in-one instrument with integrated qPCR capability that processes up to 288 samples per run with results in less than 6 hours for single-plex assays and less than 15 hours for multiplex NGS assays. The specific biomarker panel includes 220 targets for neurodegeneration detection including Abeta variants, phosphorylated tau variants, neurofilament light chain, and numerous inflammatory and neurodegenerative disease markers. Delivery must be completed within 30 days after receipt of order to the National Institute on Aging at 251 Bayview Boulevard, Baltimore, Maryland 21224, with contact Ana Patricia Amorim Gomes at patricia.amorimgomes@nia.nih.gov.

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs
PURCHASE DESCRIPTION
1. Statement of Need and Purpose: The National Institute on Aging (NIA) requires seven
(7) multiplexed precision proteomics immunoassay panel and required bulk reagents used to run the testing and analysis for neurological disease biomarkers for an in-depth study of the protein changes with neurodegeneration and how dietary interventions may modulate them.
2. Background Information and Objective:
Chronic pain is one of the most pervasive and disabling health challenges worldwide, contributing to reduced quality of life, social isolation and cognitive decline. Despite its impact, we are still lacking objective biomarkers that can capture and track underlying physiopathological processes and predict treatment response. Moreover, despite taking multiple drugs, patients still report substantial disability. Dietary interventions represent a promising therapeutic approach by modulating neuroinflammation and glial activation which underlie chronic pain. Leveraging samples from a randomized clinical trial of fatty-acid dietary interventions in chronic pain patients, we aim to identify plasma and exosome-derived biomarkers that allow monitoring of treatment response, as well as identify novel therapeutic targets to guide precision medicine interventions in older adults living with chronic pain.
Neurodegenerative disease research is complicated by a high degree of clinical heterogeneity and the prevalence of co-pathologies. The identification of clinically relevant biomarkers for neurological disorders is critical in the development of better tools for diagnosis, disease progression, and prediction of therapeutic outcomes. NIA requires a brand name or equal NULISAseq Neuro 220 Panel to provide comprehensive coverage of the hallmarks of neurodegenerative disease with highly sensitive and specific multiplexed analysis of 220 biomarkers using only 25 μL of sample, providing researchers with an automated solution to dissect disease heterogeneity, uncover co-pathologies, and identify signatures of disease prognosis, progression, and therapeutic response in clinical cohorts.
3. Generic Name of Product: Multiplexed precision proteomics immunoassay panel for neurological disease biomarkers and bulk reagents required to run the testing and analysis.
4. Salient characteristics NIA requires the following brand name or equal items:
a. NULISAseq Neuro 220 Panel. Qty. 7
b. NULISA 10X Wash Buffer. Qty. 2
