About this file
This combined synopsis and solicitation requests quotations for a Beckman Coulter Optima XPN-90 ultracentrifuge. The National Institutes of Health, National Center for Advancing Translational Sciences requires the ultracentrifuge to use patient tissues from its biobank to confirm findings are relevant to improving lives of patients. Quotations are due by February 9, 2024. Award will be made based on technical capability, past performance, and price. The firm fixed price purchase order will be made in accordance with the Prompt Payment Act. Responses must identify where parts are manufactured and indicate if the proposed product meets salient characteristics. Registration in the System for Award Management is required.
The salient characteristics include a 15-inch touchscreen, heating and cooling system without compressors, direct air cooling, noise level below 51 dBa, vacuum level below 5 microns, idle power consumption of 60W or less, compatibility with Beckman Coulter rotors and tubes, and software with export functions. Delivery within 30 days of award is requested. A three-year warranty is required, and pricing for extended service agreements up to three additional years should be provided.

PURCHASE DESCRIPTION
1. Statement of Need and Purpose: The Neurosurgical Oncology Unit uses several molecular biology and biochemistry techniques in our scientific mission to better understand pathogenesis of gliomas and develop better therapies. While we perform several assays such as cell cycle, a technology that provides rapid multi-parametric analysis of cells in soluti, etc. We do not currently have instruments within our group to efficiently and reliably do this work. The ultracentrifuge will allow us to perform several assays in a single unit and will expand the repertoire of biochemical and molecular biology techniques our group will be able to perform.
The procurement of the Beckman Optima XPN-90 Ultracentrifuge will provide intra-laboratory high content ultracentrifuge for our laboratory that will be compatible with current research protocols within the NIH and compatibility with data and software on the current ultracentrifuge research facility which provides basic research support to members as well as to investigators throughout NIH Support is provided using instruments manufactured by Beckman. Our laboratory requires this same type of instrument for seamless operation and data acquisition.
The research projects supported are complex and varied. As such, the laboratory is required to maintain equipment that provides for a wide range of applications and that includes the ability to modify and adapt capabilities as new applications are developed.
2. Project Background: Glioblastomas are the most common malignant CNS neoplasm. Despite aggressive surgery, chemotherapy and brain radiation, median survival is only 15 months and the disease remains uniformly fatal. The NOU performs mission critical research on the molecular and cellular biology unique to glioblastomas with an expressed goal of developing novel therapeutic strategies to help patients. We recently used a publicly available genetic glioma dataset to probe a novel hypothesis regarding how primary cilia may affect glioma signal transduction and cancer phenotype. The data was confirmed using a separate, unrelated, publicly dataset. The goal of this purchase through GeoMx is to use tissues from our patients available in the biobank within the Surgical Neurology Branch (SNB) to confirm that our findings are indeed relevant to the patient population here at NIH whose lives we seek to improve. This is a necessary and important step in order to publish our findings in a high-impact journal. Additionally, the information gained will allow us to leverage our limited resources to determine and develop the targets with highest success potential. • Qty 1, Optima XPN 80 installation service
Trade Name: Beckman Optima XPN90
Products brand name: Beckman Coulter Optima XPN-90
Manufacturers name: Beckman Coulter
5. Salient Characteristics Our laboratory is in need of an ultracentrifuge with user-friendly, advanced intelligence software and large touch-screen display that is easy to operate and effortlessly manages security and electronic records, increasing productivity and ensuring confidence. In addition, it is imperative that we obtain a reliable, state-of-the-art ultracentrifuge with powerful simulation tools and networking capabilities to meet our present and future needs. The ultracentrifuge will help us perform our current separations including extra cellular vesicles and develop new procedures with its innovative, intuitive software. We are unable to find an instrument from another vendor with these necessary features.
• Must have 15” LCD display with ± 45° horizontal and vertical tilt for easy operation (even with gloved hands) and viewing from virtually anywhere in the lab.
Must have unique Beckman SW32Ti Rotor Package.
• The drive must be covered by a 10-year warranty.
• Must have heating/cooling system that does not use compressors or chlorofluorocarbons (CFCs). The thermoelectric heating/cooling unit provides fast chamber/rotor refrigeration or heating and does not require any compressors, reducing instrument down time and heat and noise levels.
• Must have an air-cooled direct drive system that requires no gears, brushes, oil vacuum seal, external oil reservoir, or continuously-operating damper. This provides efficient operation and extends the life of the instrument. Air cooling eliminates chlorofluorocarbons and compressors which results in less instru-ment down time and less output of heat and noise.
• Must operate at a noise level below 51 dBa.
• Must achieve vacuum level below 5 microns (0.7 Pa).
• Must consume no more than 60W of power while idling (powered on, but not running).
• Must have available Near Vertical Tube (NVT) rotors to maximize lipoprotein and plasmid DNA separa-tions for speed and purity.
• Must be compatible with all currently-produced Beckman Coulter floor preparative ultracentrifuge rotors.
• Must have tube options including: OptiSeal (for easy sealing without tools), Quick-Seal (fast heat-sealing), and g-Max (shortens separation times by up to 50 percent) and Ultra Clear Tubes.
• Must be designed and tested as a system with Beckman Coulter rotors, tubes and accessories.
• The Contractor shall provide an Original Equipment Manufacturer (OEM) certified technician to perform the setup and installation of the equipment.
SOFTWARE
