Attachment 1 Performance Work Statement and QASP Wheat Genotyping – Raleigh NC
1. Background and Scope The United States Department of Agriculture, Agricultural Research Service, Plant Science Research Unit, Raleigh, North Carolina requires targeted genotyping-by-sequencing (GBS) of 1,920 wheat samples with the AgriSeq Wheat 4k genotyping panel. Only the AgriSeq Wheat 4k genotyping panel is acceptable for requirement performance to maintain critical continuity of research.
The AgriSeq wheat 4k genotyping panel consists of 4000 specific informative genomic targets identified by scientists at the USDA-ARS Regional Small Grains Genotyping Laboratories. The resulting genetic marker data will be used in conjunction with the Plant Science Research Unit’s investigation into the complex genetic traits of plants.
2. Privity of Contract - Contract service providers shall be accountable solely to the Prime Contractor, who, in turn, is responsible for contract performance.
3. Tasks or Requirements:
The contractor shall provide:
3.1. DNA library preparation for 1,920 wheat DNA samples for AgriSeq targeted GBS using the Wheat
4k genotyping panel with rescue primer spike-in. DNA samples will be provided by the USDA program office point of contact to the contractor. Receipt confirmation of the samples shall be provided in writing to the USDA program office point of contact.
3.2. Sequencing of prepared AgriSeq DNA libraries with a mean target coverage of no less than 100x.
3.3. Delivery of AgriSeq wheat panel target variant calls to the identified USDA program office point of contact shall be in the form of a single merged Variant Call Format (VCF) file containing call data for all submitted samples.
3.4. All deliverables, including library preparation and sequencing shall be delivered to the USDA point of contact within 30 calendar days of contractor receipt of the DNA samples from the USDA.
4. Quality Assurance Surveillance Plan (QASP). The Government shall establish a QASP as a means of evaluating whether the Contractor is meeting the performance standards/quality levels identified in the
PWS to ensure that the government pays only for the level of services received
