STATEMENT OF WORK (SOW) FOR

LIQUID CHROMATOGRAPHY-MASS SPECTROMETRY (LCMS/MS) ANALYZER AT

VA PHILADELPHIA VA MEDICAL CENTER

3900 WOODLAND AVENUE

PHILADELPHIA, PA

19104-4551

1. Contract Title.

Procurement of a Liquid Chromatography-Mass Spectrometry (LCMS/MS) Analyzer and all associated equipment, computers, Monitor, training. License, software, gas generator, accessories, and initial chemicals needed to perform Drug confirmation testing using LCMS/MS at the Michael J. Crescenz VA Medical Center.

2. Background.

Pathology and Laboratory Medicine, Chemistry and Toxicology section needs new equipment that can analyzer patient’s urine to detect and confirm drugs detected initially on a drug screen or drugs being prescribed to veteran patients to confirm the presence in random urine. This equipment will replace a 15-year-old instrument which is being used until a replacement is acquired. It is beyond the life expectancy of the analyzer. We are looking for a replacement that have minimal maintenance, prevents source contamination, possesses increased sensitivity, and significantly decreases in-source carryover to be able to detect and confirm the presence of drug analytes in human urine.

3. Requirements. PVAMC requires an LCMS/MS QTRAP System that has the following:

3.1 TurboV Ion Source: a unique heater geometry that places the desolvation heaters (x2) orthogonal to the ESI electrode. This configuration focuses heat on the ESI droplets for efficient desolvation just above the axis of the orifice which results in superior sensitivity and significantly decreases in-source carryover.

3.2 Curtain Gas interface design to consist of a neutral gas (N2) that flows out from between the curtain plate and the orifice. This orifice must be a single thin aperture separating the vacuum chamber from atmosphere. This design is different that the other interface designs that utilize a long metal tube to transfer ions from the atmosphere into the vacuum region of the MS. This design prevents neutral and other ion clusters from entering the mass analyzer and has advantages over the ion transfer tube designs that are prone to clogging.

3.3 QJET is needed to capture the ions in the free jet expansion cloud behind the orifice and to transfer them to the lower pressure regions of the mass spectrometer (Q0, Q1, Q2, Q3). The QJet Ion Guide is an RF only quadrupole that focuses the ions into the lower pressure Q0 region while efficiently removing gas molecules. The QJET coupled with the curtain gas and orifice design gives a rugged and robust advantage.

3.4 QTRAP: which is a hybrid triple quadrupole/linear ion trap mass spectrometer that can accommodate both Quantitative and qualitative LC-MS/MS workflows. QTRAP gives one the ability to use both the triple quad and ion trap scan functions on the same platform even with a single LC-MS/MS injection. In addition to MRM quant, the QTRAP system can be used to rapidly acquire MS/NS spectra for every detected peak (by MRM) that exceeds a threshold. Library searching of MS/MS spectra then allows the inambiguous identification of chromatographic peaks.
