Attachment 001 Performance Work Statement and QASP Mass Spectrometry-Based Untargeted Metabolomics

1. Background Scientists from the ERRC are evaluating the effect of different cooking oils on the metabolism of the gut microbiome as well as identifying microbial transformation of lipids.

2. Scope ERRC will use this acquisition to obtain mass spectrometry-based untargeted metabolomics results to further research goals.

3. Objectives Scientists from the DFFRU at the ERRC expect to receive untargeted metabolomics results and raw data files from this analysis. Results will be analyzed within the context of the experimental design and shared within manuscripts and presentations.

4. Tasks or Requirements:

4.1. Preliminary task by Government: ERRC scientists will ship filtered supernatant from bacterial cultures overnight on dry ice for mass spectrometry-based untargeted metabolomics and data analysis. Contractor is not responsible for shipping cost from Government to Contractor.

4.2. Preliminary task by Government: Breakdown of samples shipped by ERRC shall be as follows:

4.2.1. 45 Samples for primary metabolism, complex lipids, and biogenic amines

4.2.2. 5 QC samples

4.2.3. 6 lipids samples

4.3. Upon delivery samples will be thawed and processed for analysis.

4.4. Processed supernatants will be analyzed using three panels: primary metabolism, complex lipids, and biogenic amines. Combined these panels must be capable of providing data for over 1000 validated compounds and others of unknown chemical structure.

4.5. Data shall be reported with retention times, quantification masses, biochemical database identifiers, and adduct information. Metabolites shall be reported as relative peak heights, but absolute quantitation shall be available if requested

4.6. Metabolite annotations must be assigned a level of confidence rating (minimum 4, but ideally 5 levels), as follows:

4.6.1. Confirmed structure match based on MS, MS/MS, and RT of chemical standard.

4.6.2. Probable structure based on MS/MS match in a spectral database.

4.6.3. Tentative structure based on MS1 m/z database match.

4.6.4. Molecular formula identification based on isotope abundance, adduct formation etc.

4.6.5. Exact mass m/z.

4.7. Raw data files and methods shall be available to share with ERRC.

4.8. The total period of performance is 180 days.

4.8.1. 30 days after receipt of order will be allowed for the Government to complete Preliminary tasks.

4.8.2. The remainder of the tasks shall be completed within 150 days of sample delivery receipt.

5. Quality Assurance Surveillance Plan (QASP). The Government shall establish a QASP as a means of evaluating whether the Contractor is meeting the performance standards/quality levels identified in the

PWS to ensure that the government pays only for the level of services received
