About this file

This document is a Statement of Work (SOW) for a federal contract opportunity to acquire services for the manufacture of clinical grade lentiviral vector encoding GPC2-CAR.

The key details are:

  • The National Cancer Institute (NCI) requires services to produce and test 250 ml of purified, Good Manufacturing Practice (GMP) grade lentiviral vector using the Government-provided pWPT-CT3-EGFRt transfer plasmid.
  • The lentiviral vector will be used to engineer T-cells from neuroblastoma patients into CAR-T cells for use in a Phase I/II clinical trial.
  • The contract requires the vendor to meet cGMP manufacturing requirements, including use of certified cell lines, GMP-compliant cleanrooms, quality assurance, and stability testing per FDA guidance.
  • The vendor must complete the production and release of the lentiviral vector within 6 months of contract award.
  • Responses to the pre-solicitation notice for this sole-source requirement are due by May 15, 2024. The Government will consider any responses to determine if a competitive procurement is warranted.

STATEMENT OF WORK

1.0 TITLE

Acquisition of Services for the Manufacture of Clinical Grade Lentiviral Vector Encoding

GPC2-CAR

2.0 BACKGROUND

The Laboratory of Molecular Biology (LMB) and the Pediatric Oncology Branch (POB) at the National Institutes of Health (NIH), National Cancer Institute (NCI), Center for Cancer Research (CCR), require services for the production and subsequent stability testing of Good Manufacturing Practice (GMP) grade lentiviral (LV) vector, pWPT-CT3-EGFRt.

Neuroblastoma accounts for 8–10% of all childhood cancers and 15% of all pediatric cancer mortality. Glypican-2 (GPC2) is a cell surface protein that is expressed in nearly half of neuroblastoma cases. High GPC2 expression correlates with poor overall survival. It modulates N-myc, an oncogenic driver of neuroblatoma tumorigenesis, through Wnt signaling. Thus, GPC2 is a promising target for adoptive T-cell therapy. In this approach, T cells are taken from the patient, engineered in the laboratory into chimeric antigen receptor- (CAR-) T cells, and reapplied to the patient to fight cancer cells.

The manufactured product under this acquisition will be used to engineer T-cells of neuroblastoma patients into CAR-T cells with the ability to specifically target GPC2-expressing tumor cells. Ultimately, this product will be used in a phase I clinical trial in the Pediatric Oncology Branch at NCI under IRB001812 entitled Phase I/II Study Glypican-2–targeted Autologous Genetically Modified CT3.CD28HTM.BBζ Chimeric Antigen Receptor (GPC2CAR)- T-cell Therapy in Children and Young Adults with Recurrent or Refractory Neuroblastoma. The protocol is being submitted to the Office of Sponsor and Regulatory Oversight (OSRO)) for review and in preparation for the Investigational New Drug Application (IND) package for submission to the Federal Drug Administration (FDA).

3.0 SCOPE

This acquisition includes services to prepare cGMP (clinical) grade transfer plasmid from a transfer plasmid provided by the Government; to obtain one (1) batch of purified, GMP grade lentiviral vector, of 250 ml in total; and to perform stability studies on the lentiviral vector product.
