About this file

This document is a Purchase Description for a brand name or equal 4D Nucleofector system and component parts for the National Institute of Neurological Disorders and Stroke (NINDS). NINDS requires this system to provide precise and efficient gene editing tools, especially for applications in regenerative medicine and disease modeling, and intends to utilize it for the introduction of CRISPR Cas9 RNPs into induced pluripotent stem cells (iPSCs) to introduce neurodegeneration-related mutations. The required items include a 4D Nucleofector X Unit, Plug IEC C13 USA, and 4D Nucleofector Core Unit. The salient characteristics include supporting nucleofection of various cell numbers, featuring positions for 20 mL Nucleocuvette Strips and 100 mL single Nucleocuvette, providing seamless transfer of conditions between different Nucleofection Vessels, possessing an electrically driven drawer for cuvette retainer, and being able to efficiently deliver CRISPR-Cas9 complexes. Delivery is required 30 days After Receipt of Order (ARO) to the Ward Lab at the specified location.

PURCHASE DESCRIPTION

1. Statement of Need and Purpose:The National Institute of Neurological Disorders and Stroke (NINDS) requires a brand name or equal 4D Nucleofector system and component parts for genome engineering of human induced pluripotent stem cells (hiPSCs) using CRISPR-Cas9 technology. NINDS requires a system that will provide precise and efficient gene editing tools, especially for applications in regenerative medicine and disease modeling. NINDS requires a 4D Nucleofector that allows for precise and efficient gene editing tools, especially for applications in regenerative medicine and disease modeling. NINDS intends to utilize this system for the introduction of CRISPR Cas9 RNPs into iPSCs to introduce neurodegeneration related mutations.

2. Background Information and Objective: The Ward lab is an internationally recognized research group that uses human iPSC models to study neurodegenerative diseases.

Genome engineering of iPSCs is necessary to accurately model disease-associated mutations, leveraging CRISPR Cas9 technology.

3. Purchase Description: NINDS requires the following brand name or equal items:

Qty. Description – Brand name or Equal 1 4D Nucleofector X Unit (AAF-1003X) 1 Plug IEC C13 USA (KAA-01US) 1 4D Nucleofector Core Unit (AAF-1003B)

4. Salient characteristics:

NINDS requires a system with the following brand name or equal salient characteristics:

• Support Nucleofection of various cell numbers (2 x 104 to 2 x 107) cells in different formats.

• Feature positions for 20 μL Nucleocuvette® Strips and 100 μL single Nucleocuvette®.

• Provide seamless transfer of conditions between different Nucleofection® Vessels.

• Posses an electrically driven drawer for cuvette retainer.

• Support Nucleofection of various cell numbers in different formats.

• Allow for substrate independent transfection of DNA, RNA, RNP's and proteins.

• Provide versatility in transfection parameters, enabling optimization for different cell types including hiPSCs.

• Able to efficiently deliver CRISPR-Cas9 complexes.

• Capable of both small-scale experiments and large-scale production
