About this file

This is a Statement of Work for the PRISM CMEVALUATE System Evaluation project, a research initiative focused on developing a human multiorgan microphysiological platform to assess radiation-induced injury and evaluate candidate countermeasures. The contractor shall establish an integrated system combining cerebral organoids, cardiac organoids, liver tissues, and endothelial barriers under recirculating flow conditions to model physiological responses to ionizing radiation exposure. The work involves evaluating radiation dose-response effects at 0.5, 1, and 2 Gy exposure levels using reactive oxygen species (ROS), γH2AX/53BP1 DNA damage markers, cytokine analysis, viability assays, and tissue-specific functional assays. Additionally, the contractor must assess four candidate countermeasure compounds—NAC, Metformin, Fisetin, and Amifostine—for their effectiveness in reducing radiation-induced injury, with optional single-cell RNA sequencing available for selected groups depending on outcomes.

The work shall be performed at the Biopico Systems Inc. facility in Irvine, California over a 6-month period beginning at award, with an estimated timeframe of July 2026 through January 2027. Required deliverables include a radiation dose-response dataset, quantitative analysis of specified biomarkers and functional markers across exposure levels, evaluation results for the candidate countermeasure compounds, and a final analyzed dataset with summary report formatted to meet NASA data submission requirements.

STATEMENT OF WORK

Physiological Radiation Injury using Systemic Multiorgan and Countermeasure Evaluation (PRISM CMEVALUATE System Eval)

1. Objective/Requirements Exposure to ionizing radiation during spaceflight can cause oxidative stress, DNA damage, inflammation, senescence, and tissue dysfunction across multiple organ systems, including the brain, heart, and liver.

Current models do not adequately replicate human multiorgan interactions or vascular communication.

This project will develop the PRISM CMEVALUATE human multiorgan microphysiological platform to assess radiation-induced injury and evaluate candidate countermeasures under physiologically relevant recirculating flow conditions following 0.5–2 Gy radiation exposure.

2. Characteristics, Scope, and Specs The contractor shall perform the following duties:

• Establish a human multiorgan microphysiological system integrating cerebral organoids, cardiac organoids, liver tissues, and endothelial barriers under recirculating flow conditions to model radiation-induced tissue injury.

• Evaluate radiation dose-response effects following 0.5, 1, and 2 Gy exposure using ROS, γH2AX/53BP1, cytokine, viability, and tissue-specific functional assays.

• Assess candidate countermeasure compounds, including NAC, Metformin, Fisetin, and Amifostine, for their ability to reduce radiation-induced injury.

• Establish a functional multiorgan microphysiological system integrating brain, cardiac, liver, and endothelial compartments under recirculating flow conditions

Optional single-cell RNA sequencing may be performed on selected groups to characterize cell-type-specific molecular responses, dependent on outcomes.

The contractor shall provide the following deliverables:

• Radiation dose-response dataset following 0.5, 1, and 2 Gy exposure

• Quantitative analysis of ROS, γH2AX/53BP1 DNA damage, cytokines, viability, and tissue-specific functional markers

• Evaluation of candidate radiation countermeasure compounds including NAC, Metformin, Fisetin, and Amifostine

• Final analyzed dataset and summary report suitable for NASA data submission requirements

3. Place of Performance Biopico Systems Inc. facility in Irvine, CA

4. Period of Performance Work shall be performed over a 6-month duration beginning at award (estimated timeframe July 2026 through Jan 2027).
