About this file
This document is an amendment to a Request for Quotation (RFQ) for Neurodegenerative Disease Research - Whole Genome Sequencing, Solicitation Number 75N95024Q00297. The key details are:
The purpose of this amendment is to revise the response due date to July 12, 2024, revise responses to several questions provided in a previous amendment, and revise the Statement of Work. The SOW specifies that sequencing must be performed using the Illumina NovaSeq6000 S4 (2x150bp) or NovaSeq X Plus 25B (2x150bp) platform with Illumina's TruSeq DNA PCR-Free Library preparation kit. The contractor shall provide FASTQ raw data to a Google Cloud server within 10 weeks after samples pass initial quality control. This is a firm fixed-price purchase order expected to be awarded to a small business, with a base period of performance from August 20, 2024 to August 19, 2025 for 300 samples, and four optional one-year ordering periods for up to 4,795 samples per period.

PURPOSE OF RFQ AMENDMENT No. 2
The purpose of this amendment is to:
1) Revise response due date to July 12, 2024.
2) Revise response Questions No. 6, 7, 15 provided via Amend No.1.
3) Revise the Statement of Work – SOW (RFQ-Attachment 1)
NOTE: The SOW (page 1 – Specific requirements – 3rd sentence) is revised to read as follows:
“Sequencing shall be performed using the Illumina NovaSeq6000 S4 (2x150bp) or NovaSeq X Plus 25B (2x150bp) platform and other related instrumentation and software with 150bp paired-end sequencing method.”
GENERAL RFQ QUESTIONS/ANSWERS:
Responses to Questions No. 6, 7 and 15 provided under Amend1 are revised as shown below in bold.
Question 6: Based on the SOW, sequencing using Illumina technology is requested. Would there be an opportunity for sequencing in a long-read system using PacBio and Nanopore technologies?
Response 6: Sequencing must be performed using Illumina NovaSeq6000 S4 (2x150bp) or NovaSeq X Plus 25B (2x150bp). Alternatives are not acceptable for this requirement.
Question 7: Can sequencing be performed on Illumina Novaseq X Series?
Response 7: Sequencing must be performed using Illumina NovaSeq6000 S4 (2x150bp) or NovaSeq X
Plus 25B (2x150bp). Alternatives are not acceptable.

RFQ 75N95024Q00297 Amendment 2
Question 15: The specification requests the Illumina TruSeq DNA PCR-Free kit, and NovaSeq 6000 sequencing platform with S4 flow cell. Illumina now offers newer versions of both items that perform as well or better, and at a lower cost. Is it acceptable to offer the NovaSeq X- Plus sequencing platform with 25B flow cell, and/or the newer Illumina DNA PCR-Free kit?
Response 15: Illumina’s TruSeq DNA PCR-Free Library preparation kit (350bp) is required for this project along with NovaSeq 6000 or NovaSeq X Plus 25B (2x150bp) sequencing platform with S4 flow cell. Alternatives will not be acceptable.
