COMBINED SYNOPSIS / SOLICITATION
RFQ No.: 75N91024Q00007
Title: Acquisition of one (1) Agilent Technologies Cytation C10 Confocal
Imaging Reader or equivalent Notice Type: Combined Synopsis / Solicitation Solicitation Number: 75N91024Q00007 Posted Date: 11/27/23 Response Date: 12/7/23 (3:00 pm EST) PSC Code: 6640 – Laboratory Equipment and Supplies NAICS Code: 334516 – Analytical Laboratory Instrument Manufacturing
1.0 DESCRIPTION
This is a combined synopsis/solicitation for commercial items, prepared in accordance with the format in FAR 12.6 as supplemented with additional information included in this notice, and shall be processed in accordance with FAR Part 13 – Simplified Acquisition Procedures. This announcement constitutes the only solicitation; quotations are being requested and a separate, written solicitation will not be issued. This solicitation, 75N91024Q00007, is issued as a request for quotation (RFQ) and includes all applicable provisions and clauses in effect through FAR Federal Acquisition Circular (FAC) 2024-01 (11/14/2023), simplified acquisition procedures for commercial items.
The North American Industry Classification System (NAICS) code is 334516 – Analytical Laboratory Instrument Manufacturing and the business size standard is 1,000 employees.
One (1) firm fixed price purchase order award will be made as a result of this solicitation.
2.0 BACKGROUND
The U.S. Department of Health and Human Services (HHS), National Institutes of Health (NIH), National Cancer Institute (NCI), Center for Cancer Research (CCR), Experimental Immunology Branch (EIB) aims to understand mechanisms of normal and malignant hematopoiesis. The EIB investigates how the cellular machinery that maintains the quality of the cellular proteins safeguards and programs blood cell development and function in response to environmental stress and leukemia of blood stem cells in the bone marrow. The EIB intends to gain a better and deeper insight into how blood stem cells develop, their regenerative potential, and how diseases like acute myeloid leukemia (AML) arise. This knowledge can guide new therapies targeted at proteostasis regulators.
For these studies, the EIB routinely assays and tracks normal and leukemia blood stem cell regenerative and differentiation potential in semi-solid culture media. These assays require an automated tissue culture plate-adapted imaging instrument with which the EIB can physically, via imaging, enumerate, identify, and track phenotype by fluorescence of blood stem cell colonies. Therefore, the EIB intends to procure one (1) Agilent Technologies Cytation C10 Confocal Imaging Reader or equivalent.
3.0 TYPE OF ORDER
This is a Firm Fixed-Price Purchase Order.
4.0 SPECIAL ORDER REQUIREMENTS
4.1 REQUIRED ITEMS
The following items shall be included:
Item Product Description Quantity
Bench size multi-mode confocal fluorescent imaging unit and associated software
2 24-inch monitor, keyboard 1 3 DAPI filter 1 4 GFP filter 1 5 TEXAS RED filter 1 6 CY5 filter 1 7 Gas Controller 1
4.2 PRODUCT FEATURES/SALIENT CHARACTERISTICS
The following product features/characteristics/minimum specifications are required:
• The imaging reader shall be a benchtop instrument with a small footprint, not to exceed 27” w x 18.5” h x 20” d, in order to fit in the available laboratory space.
• The instrument shall be capable of performing the following types of experiments:
− Brightfield and fluorescence images of cells;
− Cell viability assays;
− Histological section imaging and quantification;
− Cell cycle analysis by imaging; and − Luminescence measurements
• The instrument shall meet the following imaging requirements:
− Ability to hold and image slides and multi-well (6-well, 12-well, 24-well) plates as cells are routinely cultured in these plates;
− Ability to perform brightfield and multi-color fluorescence imaging. Cultured cells express fluorescent reporters for tracking various intracellular factors and cell states;
− Shall have automated spinning disk confocal microscopy, widefield microscopy and an option for multimode detection in one system. This specification enables high-resolution and 3-dimensional (z-stack) analysis of cells;
− Shall have an available widefield inverted microscope. This is a standard option for imaging live cells;
− Shall be capable of endpoint, time-lapse, z-stack, and montage imaging methods in order to yield high-resolution 3-D images with x, y and z-depths;
− Shall have a 6-objective capacity for confocal and inverted microscope;
− Shall have 4-microscopy color channels (for confocal and widefield inverted microscope, plus brightfield, to cover the range of fluorescence utilized for EIB’s studies;
− Shall have 20x to 40x magnification with confocal microscope system, to cover broad imaging applications. These resolutions allow detailed imaging of fluorescence within cells;
− Shall have 1.25x to 40x magnification with widefield inverted microscope system, to cover broad imaging applications. These resolutions allow broad or detailed brightfield imaging of cells;
− Shall have the option for hit-picking function. This allows EIB to use the plate reader optics to automatically focus and identify samples of interest for focused imaging;
− Shall include a gas controller for monitoring and control of CO2 and O2 levels to support live cell assays. The gas controller shall use 100% CO2 gas bottles and automatically maintain 5% CO2 in the incubation chamber instead of 5% premixed gas;
− Shall be compatible with 2 μL low volume microplate accessory with 16- or 48-sample microspots for direct nucleic acid quantification as well as fluorescence and luminescence measurements. This minimizes sample volume/wasted when performing assays; and
− For consistency in data quality, parameters, metadata, and processing with EIB’s previous assays, instrument parameters (image resolution, pixel counts, z-stack settings, cell colony morphology and resolution) shall be compatible with a previously used Cytation C5 instrument.
• The instrument shall meet the following software requirements:
− Shall have available spinning disk confocal microscope with 40 and 60 μm disks in the instrument at the same time. The user shall be able to switch between the spinning disks via software selection without manual intervention;
− Shall include image analysis software for image processing, automatic cell counting and subpopulation analysis;
− Shall have data analysis software for quantitative and qualitative analysis; and − Shall have 7 log dynamic range for fluorescence measurements
