A technically acceptable quote is one in which the quoter(s) complies with the requirements in the (SON) and does not take exception, nor object to the terms of the RFQ. Quotes that are not technically acceptable will not be considered for award. The award will be made on an all or none basis.

The Government reserves the right to assess the lowest priced quote only. If the lowest priced quote is found to be technically unacceptable, the Government will then assess the second lowest priced quote and continue this process until an award can be made.

The Government intends to evaluate quotations and make an award without conducting “discussions.”

Quotes which merely offer to conduct services in accordance with the requirements of the Government's scope of work will not be eligible for an award. Therefore, the Quoter’s initial quote should contain the best terms from a price and technical standpoint. The Government reserves the right to request revisions that seek additional price discounts, revisions to the technical quote, or clarifications for aspects of the quote. If the Government seeks revisions or clarifications, the Government will provide a reasonable deadline for submission of those revisions/clarifications based on the complexity of the request. The Government reserves the right to request revisions or clarifications from only one or a select number of Quoters, if doing so would be in the public’s interest. The Government may reject any or all quotes if such action is in the public interest; accept other than the lowest quote; and waive informalities and minor irregularities in quotes received.

Prior to award the Government will require written concurrence from the contractor prior to issuance of an executed award.

1.0 TITLE

75N91024-CSS-6904112 Page 5 of 7

Meso Scale Discovery MESO QuickPlex SQ 120MM Reader for performing immunoassays.

2.0 BACKGROUND

The Laboratory of Molecular Biology (LMB) conducts both preclinical and clinical studies developing antibody-based agents, such as immunotoxins, for the treatment of cancer. This research often requires measuring the amounts of these antibody-based agents in the serum or plasma of experimental animals or patients who are receiving treatment in the NIH Clinical Center. One ongoing project in LMB is studying the molecular response of Hairy Cell Leukemia (HCL) and Hairy Cell Leukemia variant (HCLv) patients to the immunotoxin Moxetumomab pasudotox in order to understand mechanisms of treatment sensitivity and resistance. Resistance is often seen in conjunction with the development of antidrug antibodies (ADAs). Therefore, these studies require the measurements of both the immunotoxin drug and ADAs in patient blood.

Immunoassays are used to measure antibodies and antibody-based agents in various fluid samples, including serum and plasma. It is advantageous to use detection systems that are sensitive (able to detect very low levels of antibody), but it is also important that a wide range (from very low to very high) of antibody concentrations can be detected from the same sample.

Finally, it is useful to detect more than one type of antibody in the same sample, avoiding multiple separate assays (especially for precious/limited patient samples).

Electrochemiluminescence Detectors can fulfill each of these three functions.

2.1 BRAND NAME

The MESO QuickPlex SQ 120MM Reader is manufactured and sold solely by Meso Scale Discovery (MSD), a division of Meso Scale Diagnostics, LLC. of Rockville, MD. The instrument is compatible only with microwell plates and immunoassay kits sold by MSD and vice versa. MSD utilizes electrochemiluminescence to detect binding events on patterned arrays including protein drugs and antibodies. There are no other known technologies that enable multiplexed measurement using electrochemiluminescence for detection. There are no other known suppliers that provide an electrochemiluminescence detection system that is not reliant on beads or flow cells, which have a more restricted range of detection. The MSD platform is unique in its method for detection. MSD’s ECL detection utilizes proprietary SULFO-TAG™ labels that emit light upon electrochemical stimulation initiated at the electrode surfaces of MULTI-ARRAY® and MULTI-SPOT® microplates through biomolecular interactions between the analyte and labeled reporter at the surface.

2.2 OBJECTIVE

The MESO QuickPlex SQ 120MM Detector will be used for studying the molecular response of HCL and HCLv patient leukemic cells to Moxetumomab pasudotox treatment to understand mechanisms of treatment sensitivity and resistance. Resistance is often seen in conjunction with the development of antidrug antibodies (ADAs). Therefore, these studies require the measurements of both the immunotoxin drug and ADAs in patient blood. This instrument will also be used to measure the levels of antibody-based immunotoxin and ADAs in clinical samples from HCL and HCLv patients.
