About this file
This is a Request for Quote (RFQ) issued by the Food and Drug Administration (FDA) for an integrated digital platform for lyophilization research. The solicitation seeks a comprehensive digital system that integrates process data, process analytical tools (PATs), and digital twin technology for a laboratory freeze-dryer in a non-GMP environment. The primary objective is to enhance FDA's capability to conduct advanced lyophilization research, train workforce on emerging manufacturing technologies, support product quality research, enable real-time process optimization, and advance quality-by-design principles for lyophilized drug products.
The digital platform must perform closed-loop nonlinear model predictive control, automatically execute integrated control sequences, maintain 99% uptime, and support real-time data acquisition from various sensors and analytical tools. Key technical requirements include estimating process parameters with high accuracy, monitoring critical process parameters, integrating process analytical technologies, and providing digital twin capabilities with bidirectional information flow. The contract is a firm fixed-price award using a lowest price technically acceptable (LPTA) evaluation method. Delivery will be to the FDA White Oak Campus in Silver Spring, MD, with installation, on-site training for six scientists, and a one-year warranty period.

Request for Quote
Statement of Work
Integration platform of PATs and digital twin for Lyophilization
This is a combined synopsis/solicitation for commercial products or commercial services prepared in accordance with the format in subpart 12.6, as supplemented with additional information included in this Request of Quotation. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued.
1. INTRODUCTION
1.1 Background
The FDA promotes advanced manufacturing technologies in pharmaceutical production because these technologies can make manufacturing processes more reliable and strengthen supply chains. This leads to faster access to quality medicines for Americans. Digital twins and process analytical technologies (PATs) are key components of this digital transformation. These tools enable process automation, optimization, and performance prediction through real-time data analysis.
The Division of Product Quality Research conducts research on advanced manufacturing to speed up the review of advanced manufacturing technologies and support the adoption of new lyophilization technologies. This acquisition directly supports the Agency's mission to modernize the U.S. pharmaceutical industry. The equipment will enable FDA staff to better understand and identify risks to product quality and patient safety related to digitalization and advanced lyophilization technologies and accelerate regulatory assessment of manufacturing processes that use advanced process analytics. This purchase supports the requirements of the Biosimilar User Fee Act (BSUFA), Prescription Drug User Fee Act (PDUFA), and Generic Drug User Fee Act (GDUFA) programs.
