Requirement:

The Food and Drug Administration (FDA), the Office of Pharmaceutical Quality Research (OPQR) requires two (2) complete benchtop bioreactor system capable of cultivating Escherichia coli with process monitoring and automated control for temperature, pH, dissolved oxygen, foam levels, and turbidity. This system will be used to investigate the impacts of cell extract preparation on final product quality outcomes during the commercial manufacture of human therapeutic proteins using CFPS platforms. Cell-free protein synthesis (CFPS) is the in vitro expression of genes into proteins. Although CFPS has existed for more than 60 years, the application of the CFPS platform to commercial manufacture of human therapeutic proteins is new, only emerging in recent years. As an emerging manufacturing technology, the criticality of process parameters and process controls on final product quality is not yet well characterized and stakeholders lack formal guidance from regulatory agencies. One critical step in the CFPS workflow is the preparation of cell extracts that provides the necessary translational and transcriptional machinery (e.g., ribosomes, tRNAs, etc.) required for in vitro protein synthesis outside of the living cell. These extracts, also known as the “chassis” of CFPS, most commonly come from microbial hosts such as Escherichia coli but have been made from other cell types including mammalian, insect, plant, and yeast.

Background:

The FDA/OPQR requires two (2) complete benchtop bioreactor system capable of sustaining and controlling a high-density E. coli culture in a 10 L working volume. The system should be comprised of a central control tower installed with software that enables continuous process monitoring and fully automated control of key process variables: temperature, dissolved oxygen, pH, foam, and turbidity. At a minimum, the tower should support the connection, calibration, and real-time data reading for the following in-process sensors: temperature, pH (analog), DO (digital/optical), foam, and turbidity (light back scatter). However, the system should be capable of accepting additional sensors that may be desired in the future, such as for viable biomass and weight. Agitation and mixing should be driven by a bi-directional, direct drive system using a stir shaft equipped with two Rushton-type, 6-blade impellers suitable for microbial culture and a compatible stainless baffle insert. The control tower should feature individual built-in Mass Flow Controllers (MFC’s) for four each separate gas lines (e.g. nitrogen, oxygen, air, and carbon dioxide). The control software should feature manual and automated PID control over gas mixing. The system must have at least one onboard MFC to control sparge into the vessel and capable of flow rates up to 2 SLM or 20,000 ccm. A ring-type sparger should be used, and the vessel exhaust should be equipped with a condenser/exit gas cooler with flexible joint. Control of DO/pO2 levels should be automatic and be able to leverage changes in stirrer speed, gas volume flow rates, and percent oxygen in the gas mix, or all three in a cascade, if desired. Vessel temperature control must be able to leverage both heating and cooling (e.g., water-jacketed vessel supplied by a recirculating chiller) and not rely on an external, fresh water source. The control tower should be equipped with at least four variable speed, bi-directional peristaltic pumps to support the automated addition of up to four substrates to control pH (acid and base), foam (anti-foam), and Feed/Level. Two (2) sets of probes and inline sensors required for precise bioprocess monitoring and control should be included, one for each system: pH, dissolved oxygen (DO), temperature, filling level, foam, and a non-invasive sensor capable of online biomass monitoring of E. coli cultures that correlates well with offline OD600 measurements. The system should come with all necessary probes, fittings, accessories, cables, hosing, connectors, software, and any other items that would fulfill the requirement for a complete bioreactor system that can be used upon set-up for the controlled cultivation of E. coli. More details are provided below in Section 3: Technical Specification.
