STATEMENT OF WORK
For Quantitative Modeling and Simulation Approaches for Application in the Development and Approval of Generic Drug Products (2-3 day) Training
1. Introduction The Center for Drug Evaluation and Research (CDER) performs an essential public health task by making sure that safe and effective drugs are available to improve the health of the American public. As part of the U.S. Food and Drug Administration (FDA), CDER regulates Over the Counter and prescription drugs, including generic drugs.
The Office of Generic Drugs (OGD) within CDER is responsible for the approval of generic drug applications submitted to the Agency. Within OGD, the Office of Bioequivalence (OB) is responsible for the scientific assessment of bioequivalence data submitted in support of said applications. OB staff evaluates formulations for quantitative and qualitative (Q1/Q2) equivalence, reviews all Pharmacokinetic (PK) and Pharmacodynamic (PD) studies submitted with applications, and collaborates with Office of Research and Standards (ORS) and Division of Clinical Review (DCR) to consider newer methodologies from sponsors for demonstration of equivalence in complex dosage forms while ensuring integration and synchronization with the Division of Clinical Safety & Surveillance (DCSS) on products that have been identified as having potential safety or therapeutic issues.
Quantitative Modeling and Simulation (M&S) approaches include physiologically based pharmacokinetic (PBPK) models for locally acting and systemically working drug products, population pharmacokinetic (PopPK) modeling, pharmacokinetic and pharmacodynamic (PK/PD) modeling, and empirical approaches. For generics, these models can be used within the scope of alternative bioequivalence approaches, product specific guidance document development, and supportive evidence for drug approval among other areas.
2. Background OB requires professional services support to facilitate a 2-3 day quantitative modeling & simulation approaches training to ensure staff are well prepared to address the utility of these approaches in generic drug development and approval. The training course will cover concepts and application of PBPK modeling, popPK modeling, PK/PD modeling, and empirical approaches for the development and approval of generic drug products.
Using its specialized knowledge and skills, the contractor will facilitate 2-3 day in-person course with recorded components. Their in-person sessions are designed for student/facilitator interaction.
3. Purpose The purpose of this requirement is to increase OB assessor and technical staff’s knowledge, skills, and abilities to perform their duties in assessing regulatory submissions containing quantitative modeling approaches to support the approval of their generic drug product. These approaches can contribute to the totality of understanding of a drug and can further inform the agency in regulatory decision-making. The agency has spearheaded several efforts to support this thinking including FDA’s Modeling-Integrated Evidence Pilot Program for Generics and CDER’s New Quantitative Medicine Center of Excellence. This course will provide a comprehensive overview of the principles, techniques, and applications of PBPK modeling, popPK modeling, PK/PD modeling, and empirical approaches with an emphasis on bioequivalence (BE) for generics. Lectures and examples from the literature will be provided with course notes for all participants. Selected models or examples will include computer code to show how to quantitate typical experimental data. At the conclusion of this course, the assessors with basic knowledge in pharmacokinetic will be able to understand the diverse array of available PBPK modeling, popPK modeling, PK/PD modeling, and empirical approaches and begin to apply them to experimental data and simulate anticipated drug responses.
Specific topics that will be considered in this training include the following:
| 3.1. | Quantitative M&S approaches to reduce/replace in vivo BE studies (i.e., in vivo PK BE studies, comparative clinical endpoint or pharmacodynamic endpoint BE studies) for complex generic products and other generic products with clinical study challenges including long acting injectables and implantable products, oncology drug products, and orphan drug products. |
| 3.2. | Quantitative M&S approaches for modified release solid oral products to ensure the consistency and quality of BE recommendations. |
| 3.3. | Quantitative M&S approaches for narrow therapeutic index drugs, long acting injectables, and complex drug products and clinical trial simulations to aid risk-based BE evaluation. |
| 3.4. | Quantitative M&S approaches to determine the appropriate study design and evaluate sensitivity for BE assessment. |
| 3.5. | Quantitative M&S approaches with consideration of condition of use (taken with food), special populations (pediatrics, elderly), sex (male a/or female) based on safety profile of innovator drug product. |
| 3.6. | Quantitative M&S approaches for biowaivers or waiver of in-vivo BE studies. |
| 4. | Objectives |
The objective of the Quantitative Modeling & Simulation Approaches two (2) to three (3) day training course is to provide CDER/OGD/OB staff a training/educational opportunity to help enhance their skills and abilities to grow in the area of Pharmacometrics which provides an understanding of the quantitative M&S approaches in regulatory assessment.
5. Scope The purpose of this procurement is to seek the full professional training and facilitation services necessary to conduct the “Quantitative Modeling and Simulation Approaches for Application in the Development and Approval of Generic Drug Products “ to OB staff members. The 2-3 day course on concepts and applications of quantitative M&S approaches will be presented to fifty (50) OB staff members. The contractor shall provide the following:
| 5.2. | Quantitative M&S approaches for modified release solid oral products to ensure the consistency and quality of BE recommendations. |
| 5.3. | Quantitative M&S approaches for narrow therapeutic index drugs, long acting injectables, and complex drug products and clinical trial simulations to aid risk-based BE evaluation. |
| 5.4. | Quantitative M&S approaches to determine the appropriate study design and evaluate sensitivity for BE assessment. |
| 5.5. | Quantitative M&S approaches with consideration of condition of use (taken with food), special populations (pediatrics, elderly), sex (male a/or female) based on safety profile of innovator drug product. |
