Requirement and Background

The US FDA’s National Center for Toxicological Research (NCTR) requires a PM and unlimited repair service contract on a (Molecular Devices) ImageXpress Micro Confocal (IXMC) (Ser #:

5150127) High Content Imaging System for a Base plus four option years to ensure system functionality for multiple ongoing projects with the Division of Systems Biology (DSB). The instrument was purchased in 2022 and was covered under the original manufacturer’s warranty which expired 11/16/2023; a service contract is required as soon as is possible.

The National Center for Toxicological Research (NCTR) has ongoing studies that rely on the capabilities of the existing ImageXpress Micro Confocal (IXMC) High Content Imaging System. A service contract on the instrument will allow for continual and reliable technical and engineering support to ensure functionality of the system to acquire high content imaging and other endpoint assays of alternative cell models for toxicity critical to on-going multiple projects with the Division of Systems Biology (DSB), such as the following studies:

Project 1: Imaging in liver microphysiological system to establish cellular assay to predict cytotoxicity and metabolic activity.

NCTR is investigating drug-induced liver injury (DILI) in a liver microphysiological system (MPS) to evaluate the physiological relevance and clinical predictivity of the system. This MPS is advantageous in that the design allows primary human hepatocytes to be co-cultured with liver non-parenchymal cells consisting of sinusoidal endothelium, Kupffer, and stellate cells in two overlapping cell culture compartments separated by a porous flexible polymer membrane. The co-culture more closely mimics in vivo functionality and physiology. Various cytotoxicity and metabolic assays of interest are being utilized that are available for traditional in vitro studies of DILI, but very few are optimized for this MPS. NCTR is interested in establishing a fluorescence-based optical detection method for effective cellular analysis of protein expression, metabolism, oxygen/glucose concentration and viability.

However, due to the three-dimensional architecture of the MPS, the flexible polymeric material used in the system and the fact that cells are cultured in an overlapping condition it is quite difficult to image with a traditional widefield microscope system. The ability to capture images using either widefield or confocal modalities would better enable imaging of the cells in the MPS.

Project 2: Ca2+ imaging with Engineered Heart Tissue

The combination of contractile force analysis along with of cytoplasmic changes in calcium concentration has the potential to provide important insights into the physiology or pathophysiology.

In vitro investigation of changes in calcium flux has been well established with invasive calcium dyes.

At NCTR, we are evaluating the application of heart tissue generated from human induced pluripotent stem cells genetically modified with calcium indicator for investigating cardiac functions and cytotoxicity when challenged with anthracycline. In our study, we proposed to non-invasively measure the calcium transient along with the force of contraction. To capture such phenomena in vitro, the DSB requires a highly efficient imaging system.

The PM and unlimited repair service contract shall consist of a Base year plus four (4) possible one-year options. Each year shall be funded at the time the option year is exercised; options will be exercised based on funds availability and the needs of the Federal Government. The initial Service period shall be for one year and the subsequent periods of performance may be renewed at the government’s option for the period between 12-24 months, 24-36 months, 36-48 months and 48-60 months beyond the initial contract service period.

All services required are as noted in the Minimum Performance Requirements Section contained herein.

Contractor Performance Evaluation(s) In accordance with FAR Subpart 42.15, FDA will complete annual (interim) and final contractor performance evaluations. Annual evaluations will be prepared to coincide with the anniversary date of the contract. Additional interim performance evaluations may be prepared at Contracting Officer discretion, as necessary. Final performance evaluations will be completed upon contract expiration.

FDA will utilize the Contractor Performance Assessment Reporting System (CPARS) in order to execute annual and final contractor performance evaluations. CPARS is a secure Internet website located at http://www.cpars.csd.disa.mil/cparsmain.htm. FDA will register the contractor in CPARS upon receipt of the name and email address of two (2) individuals who will be responsible for serving as the Contractor’s primary and alternate CPARS contacts. Once FDA registers the contractor in CPARS, the Contractor will receive an automated CPARS email message which contains User IDs and instructions for creating a password.

Once a performance evaluation is issued, the Contractor’s primary and alternate CPARS contact will receive an email instructing them to logon to CPARS in order to review the performance evaluation.

The Contractor has 14 days from the date of performance evaluation issuance in which to review the evaluation. If the Contractor is in agreement with the performance evaluation outcome, the evaluation becomes final. Should the Contractor be in disagreement with the performance evaluation outcome, rebuttal comments must be submitted via the CPARS within 14 days from date the evaluation was issued by the FDA. Any disagreement between the Contracting Officer and the Contractor will be referred to a contracting official one level above the Contracting Officer, whose decision will be final.

Copies of each performance evaluation and contractor responses, if any, will be retained as part of the official contract file and will be used to support future award decisions. Evaluations will also be stored for a 3-year period in the https://www.cpars.gov/ system.

Contractors may obtain CPARS training material and register for on-line training at https://www.cpars.gov/lc.htm . There is no fee for registration or use of the CPARS.
