The U.S. Food and Drug Administration (FDA) has a need to procure one (1) liquid chromatograph (LC) coupled to a gas chromatograph (GC) for the analysis of mineral oil hydrocarbons (MOH) and sterols in fats and oils.

**************************BRAND NAME OR EQUAL***************************

A. BACKGROUND AND OVERVIEW

The Office of Regulatory Science, Division of Bioanalytical Chemistry requires the purchase of an LC coupled to a GC (LC-GC) for the analysis MOH and sterols in fats and oils. Mineral oil hydrocarbons comprise a wide range of chemical compounds obtained mainly from petroleum distillation and refining. They are categorized into two main groups referred to as MOSH (Mineral Oil Saturated Hydrocarbons) and MOAH (Mineral Oil Aromatic Hydrocarbons). The system is expected to perform method DIN 16995/DGF C-VI 22 in a single chromatographic analysis. When analyzed by LC-GC, at first MOSH and MOAH are isolated by LC in two different liquid fractions which are quantitatively transferred to the head of two independent GC capillary columns located in the GC oven.

The liquid solvent is evaporated, then the isolated MOSH and MOAH are separated in parallel by GC and detected by flame ionization detection (FID). The automated processing of the two chromatograms using a dedicated software provides accurate measurement of these analytes. The addition of selected automated analytical steps increases the accuracy of the analytical results and lowers the limit of quantification. The LC-GC platform should be capable of performing automated epoxidation of unsaturated olefins, to prevent their elution in the MOAH fraction. It should also be capable of removing biogenic alkanes from the MOSH fraction by elution through an aluminum oxide column.

The analysis of MOH requires the saponification of lipids and isolation of unsaponifiable matter prior to instrumental analysis. Saponification, liquid-liquid repartitioning, and subsequent centrifugation to split liquid phases, may be performed using a robotic autosampler. In addition, after minor reconfiguration of the instrumentation according to manufacturer directions, the system must be capable of analyzing the sterols present in fats and oils. For this analysis at first the oil or fat sample is hydrolyzed using the robotic autosampler and the sterols are extracted by liquid-liquid partitioning. A portion of isolated unsaponifiable matter is injected into the LC column, and the eluent fraction containing the delta 5/delta 7 sterols is transferred to the head of a GC capillary column. Sterols are successively eluted from the GC column and measured by FID. The system must be delivered ready to be used for the analysis of MOH, and with all components and conversion instructions necessary for its use for sterol analyses. The manufacturer must demonstrate onsite that the delivered instrumentation can perform accurate MOH quantifications by analyzing onsite selected reference materials.

B. OBJECTIVES/PURPOSE

The requested equipment will replace an existing aged gas chromatograph system that is not capable of performing LC-GC analysis of MOH. The salient characteristics are summarized in the Specific Tasks.

In brief, the requested LC-GC equipment shall:

• Be capable of isolating by LC the MOSH and MOAH contained in an oil or fat sample, automatically transfer them to the head of two GC columns, provide parallel quantification of MOSH and MOAH using FID detection.

• Be delivered ready to perform MOSH and MOAH analyses according to method DIN 16995/DGF C-VI 22 in a single chromatographic analysis.

• Be furnished with a kit that permits the setup of the system for sterols analysis without the need for optimizations or purchase of other components.

• Include an HPLC with ultra-violet (UV) detection, an interface transferring analytes from LC to GC, a GC, a robotic autosampler, an extension kit for the epoxidation of olefin prior to LC separation, an extension kit for the aluminum oxide purification post LC of MOSH, a controller unit, a software to collect data from the two parallel GC separations, a software to process the MOSH and MOAH GC-FID chromatograms, and a fraction collector.

• Be managed by a single controller unit capable of operating the entire system, which shall be furnished with pre-loaded methods for MOSH/MOAH and sterol analyses.

• Be capable of performing automated epoxidation of olefins prior to the LC separation.

• Be capable of performing purification of the MOSH fraction from biogenic olefins by elution through an aluminum oxide column.

• Be capable of performing saponification of lipids and isolation of unsaponifiable matter using the robotic autosampler.

• Be capable of collecting fractions for analysis with other techniques as GCxGC-MS.

A computer system meeting of exceeding the technical requirements stated by the instrument vendor will be provided by FDA.
