Requirement and Background

The US FDA’s Center for Biologics Evaluation and Research (CBER), Office of Vaccine Research and Review (OVRR), Division of Viral Products (DVP) requires a new Thermo Fisher Scientific Tundra Cryo-Transmission Electron Microscope (TEM), 100kV, or equal, plus four (4) years of post-warranty maintenance.

The Division of Viral Products (DVP) Electron Microscope (EM) facility is designed to facilitate structural biology studies by Division and CBER investigators. The current transmission electron microscope was purchased in 2012, was originally situated at the Nicholson Lane Research Facility, and was moved to the White Oak campus during the 2014 transition. However, this unit is no longer operational and is outdated in terms of market standards for current EM instruments.

Cryo-transmission electron microscopy (cryo-EM) has emerged as an advantageous approach to understanding the 3-dimensional structure of complex proteins. For example, this was demonstrated at the earliest stages of the COVID-19 pandemic when several cryo-EM studies were completed in record time to reveal the structure of the SARS-CoV-2 Spike protein and the benefit of using targeted mutations to stabilize its 3-D structure, in order to enhance the neutralizing antibody response to candidate vaccines. Subsequently, cryo-EM has been used to characterize the Spike protein of SARS-CoV-2 variants. This technology continues to inform the lab’s understanding of how variants differ in their transmissibility and how they evade antibodies elicited to earlier viruses and vaccines. This powerful methodology is likely to continue to be a critical tool for obtaining structural information about vaccine candidates, as well as proteins that are targeted by vaccines or therapeutics.

Structural biology has long been a strength of the CBER research program. In addition to TEM and SEM capabilities, CBER research capabilities include state-of-the-art Mass Spectroscopy and Nuclear Magnetic Resonance laboratory programs. The addition of cryo-EM instrumentation would further strengthen CBER’s research capabilities into structural biology.

This requirement is for a Brand Name or Equal solution. Proposed equal needs to meet all of the Technical Requirements listed below to be deemed technically acceptable.

Salient physical, functional, or performance characteristics that “equal” products must meet:
