The U.S. Food and Drug Administration (FDA) requires the following: Glycoworks Rapifluor Mass Spectrometer (MS) N-Glycan Kits, and Supplies
***************************BRAND NAME OR EQUAL**************************
A. BACKGROUND AND OVERVIEW
The Office of Biotechnology Products (OBP) performs regulatory research work that focuses on understanding how upstream and downstream processing changes in biomanufacturing operations can affect product quality of therapeutic protein products. Additionally, OBP research seeks to understand how novel process analytical technologies (PAT) can either detect or predict product quality attributes that can affect drug efficacy or safety. Of the existing and upcoming therapeutic proteins on the market, monoclonal antibodies (mAbs) are the most common. A well-established critical quality attribute (CQA) known to affect mAb product efficacy and safety is N-glycosylation, which is typically measured using mass spectrometry. However, the detection and quantification of N-glycans requires a robust method to both separate and label the various N-glycan species in the sample. GlycoWorks RapiFluor-Mass Spectrometer(MS) N-Glycan Kit’s unique chemical attributes provide increased fluorescence quantification and supreme mass spectral response. The RapiFluor-MS N-Glycan Kit enables the use of a single label that provides valuable information from characterization to routine monitoring. This means that N-glycan analysis is much easier and more sensitive compared to other labeling methods.
OBP requires GlycoWorks RapiFluor-MS N-Glycan Kits and associated supplies to facilitate ongoing and future research endeavors and understanding how novel process developments and technologies affect mAb product quality.
System Requirements and Deliverables:
Glycoworks RapiFluor-MS 96-sample N-Glycan Kit’s unique chemical attributes provide increased fluorescence quantification and supreme mass spectral response. The kit enables use of a single label that provides valuable information from characterization to routine monitoring. This means that Glycan analysis is much easier and more sensitive. Several accessories are required to be used in conjunction with the N-glycan labeling kits to ensure optimum and accurate performance of the N-glycan analysis. These include: a dextran RapiFluor-MS Calibration Ladder, which is a standard used to ensure the mass accuracy of N-glycans detected during mass spectrometric analysis; a Glycan RapiFluor-MS Performance Test Standard, which is a performance test for UHPLC, which is the method used to separate N-glycan species before detection; an ammonium formate solution, which is added to the mobile phases of UHPLC to ensure better resolution of glycan peaks for detection; an Acquity UPLC BEH Glycan column used to separate the glycan species; an intact mAb mass check standard to assess the labeling efficiency and be used as a mass standard, and lastly, amber glass vials, inserts, and caps for the prepared glycan samples.
Glycan analysis kits must include options for two independent analytical glycan workflows that allow scientists to characterize and monitor glycan profiles using both optical (fluorescent) and/or MS (MS) data generated within an integrated UPLC/FLR/QTof MS system. These workflows enable researchers to assign and quantify low abundance glycan structures with higher confidence. Furthermore, analysis must support the detection of released glycan FLR with the option of mass spectrometry confirmation for the generation of quantitative glycan profiles with high confidence through accurate mass conformation. RapiFluor is a compound that supports the identification and mass confirmation specified in a library of Glycan Units installed within our already established waters_connect software. Therefore, we require the RapiFluor compound as the glycan labeling reagent, alongside previously described kit reagents, to accurately identify and quantity labeled glycans in data analysis and processing.

(i) Taxes are considered delinquent if both of the following criteria apply:
(A) The tax liability is finally determined. The liability is finally determined if it has been assessed. A liability is not finally determined if there is a pending administrative or judicial challenge. In the case of a judicial challenge to the liability, the liability is not finally determined until all judicial appeal rights have been exhausted.
(B) The taxpayer is delinquent in making payment. A taxpayer is delinquent if the taxpayer has failed to pay the tax liability when full payment was due and required. A taxpayer is not delinquent in cases where enforced collection action is precluded.
(ii) Examples.
(A) The taxpayer has received a statutory notice of deficiency, under I.R.C. §6212, which entitles the taxpayer to seek Tax Court review of a proposed tax deficiency. This is not a delinquent tax because it is not a final tax liability. Should the taxpayer seek Tax Court review, this will not be a final tax liability until the taxpayer has exercised all judicial appeal rights.
(B) The IRS has filed a notice of Federal tax lien with respect to an assessed tax liability, and the taxpayer has been issued a notice under I.R.C. §6320 entitling the taxpayer to request a hearing with the IRS Office of Appeals contesting the lien filing, and to further appeal to the Tax Court if the IRS determines to sustain the lien filing. In the course of the hearing, the taxpayer is entitled to contest the underlying tax liability because the taxpayer has had no prior opportunity to contest the liability. This is not a delinquent tax because it is not a final tax liability. Should the taxpayer seek tax court review, this will not be a final tax liability until the taxpayer has exercised all judicial appeal rights.
(C) The taxpayer has entered into an installment agreement pursuant to I.R.C. §6159. The taxpayer is making timely payments and is in full compliance with the agreement terms. The taxpayer is not delinquent because the taxpayer is not currently required to make full payment.
(D) The taxpayer has filed for bankruptcy protection. The taxpayer is not delinquent because enforced collection action is stayed under 11 U.S.C. §362 (the Bankruptcy Code).
